Direct Shoot Formation of Beta vulgaris L. var cicla L. and Endogenous Hormones Analysis of Regenerated Plantlet with Stem Elongation

摘  要:红叶甜菜的茎尖外植体在含有BA15.0 mg.L-1的MS培养基上生长约45 d时,在膨大叶片的边缘可见不定芽的直接产生。石蜡切片观察结果显示,该过程为典型的器官发生途径。将不定芽切割后在相同配方的培养基上继续培养,绝大部分组培苗表现出和野生型一致的表型,呈莲座叶;个别组培苗的茎伸长,呈螺旋叶序。HPLC检测结果显示,正常组培苗和茎伸长组培苗的内源生长素和赤霉素含量基本无差异。Adventitious shoots were directly formed from expanded leaf of shoot tip explants of red leaf beet(Beta vulgaris L.var cicla L.) when these explants were cultured on Murashige and Skoog(MS) medium with 15.0 mg·L-1 6-benzylaminopurine(BA) for about 45 days.The results of paraffin sections demonstrated that the adventitious shoots were formed through organogenesis.When the adventitious shoots were subcultured on the same medium as the above,the most regenerated plantlets showed the same phenotypes like rosette leaves as the wild type.While the stem was elongated in several regenerated plantlets and showed spiral phyllotaxis.The results of high-performance liquid chromatography(HPLC) analysis showed that the level of endogenous IAA and GA in regenerated plantlets with normal phenotypes and elongated stem respectively was very lower and shared little difference.
