摘  要:采用正交试验研究了NAA、6-BA对不同熟性甘蓝品种子叶期不同部位培养过程中愈伤组织诱导率的影响,同时以MS+NAA(0.4~0.7 mg/L),6-BA(2.0~4.0 mg/L)为丛生芽诱导培养基,研究了愈伤组织形态特征与丛生芽发生之间的关系。结果表明,晚丰和中甘十一的带柄子叶、不带柄子叶、下胚轴易诱导出愈伤组织;京丰甘蓝的下胚轴易诱导出愈伤组织,早熟品种下胚轴诱导的愈伤组织丛生芽发生率高于其他品种的愈伤组织。在3种甘蓝子叶期外植体培养中选下胚轴作为培养材料诱导获得的愈伤组织易发生丛生芽,京丰的下胚轴在NAA0.45 mg/L,6-BA 3.0 mg/L时诱导出中间绿色周围半透明松脆柔软团状生长快的愈伤组织丛生芽发生率达到79%;晚丰下胚轴在NAA0.6 mg/L,6-BA 6.0 mg/L时诱导出淡黄色至淡黄绿色松脆柔软团状生长快的愈伤组织丛生芽发生率可达94%;中甘十一的下胚轴在NAA0.45 mg/L,6-BA3.0mg/L时诱导出的中间为绿色周围为半透明松脆柔软团状生长快的愈伤组织丛生芽发生率可达95%。This paper was to investigate the effect of orthogonal combinations of NAA and 6-BA on callus induction of Brassica,and the relation between callus morphology and occurrence of multiple shoots under the shoot induction medium of MS+NAA(0.4-0.7 mg/L) 6-BA(2.0-4.0 mg/L).The results showed that callus is easy induced from cotyledon with petiole,cotyledon without petiole,hypocotyl of Wanfeng and Zhonggan 11,and from hypocotyl of Jingfeng;The callus induced from hypocoty is easy to occur multiple shoots.The rate of multiple shoots occurrence in callus induced from early maturity cabbage's hypocotyl is higher than the others.The rate of multiple shoots occurrence reached 79% induced by callus of middle green around semi-transparent,crisp soft,conglomerata,grow fast induced from hypocotyl of Jingfeng in MS+NAA0.45 mg/L,6-BA 3.0 mg/L;the rate of multiple shoots occurrence reached 94% induced by callus of Pale yellow to yellowish green,crisp soft,conglomerata,grow fast induced from hypocotyl of Wanfeng in MS+NAA 0.6 mg/L,6-BA 6.0 mg/L;the rate of shoots occurrence reached 95% induced by callus of middle green around semi-transparent,crisp soft,conglomerata,grow fast induced from hypocotyl of Zhongganshiyi in MS+NAA0.6 mg/L,6-BA 6.0 mg/L.
