摘 要:目的研究环境雌激素双酚A(bisphenol A,BPA)对3种激素相关肿瘤细胞株———人卵巢癌3AO细胞、前列腺癌DU145细胞及子宫内膜癌HHUA细胞增殖活性的影响。方法分别用1‰无水乙醇(溶剂对照)、BPA(终浓度为10-9mol/L)和β-雌二醇(终浓度为10-7 mol/L)染毒3AO、DU145及HHUA细胞。采用四甲基偶氮唑盐(MTT)法检测细胞(细胞密度分别为1 000、3 000、5 000个/孔)的增殖活性,并采用逆转录-聚合酶链反应检测细胞增殖因子Ki67、增殖细胞核抗原(PCNA)的mRNA表达;采用电化学发光法检测糖类抗原15-3(CA15-3)在HHUA细胞培养上清液中的含量。结果与溶剂对照组比较,BPA组3AO、DU145及HHUA细胞的吸光度(A)值较高,差异均有统计学意义(P<0.05);对3AO、DU145及HHUA细胞的促增殖率分别为10.15%,14.04%及21.31%。β-雌二醇对DU145及HHUA细胞的促增殖率分别为14.91%及9.07%,而对3AO细胞未呈现促增殖活性(P>0.05)。与溶剂对照组比较,BPA染毒组3种细胞Ki67、PCNA mRNA的表达均较高,差异有统计学意义(P<0.05);β-雌二醇染毒组DU145、HHUA细胞Ki67、PCNA mRNA的表达均较高,差异有统计学意义(P<0.05),而3AO细胞Ki67、PCNA mRNA的表达与溶剂对照组比较,差异无统计学意义。与溶剂对照组比较,BPA染毒组和雌激素对照组HHUA细胞CA15-3分泌量均较高,差异有统计学意义(P<0.05)。结论 BPA可以促进3AO、DU145及HHUA细胞的增殖活性。Objective To study the effect of bisphenol A (BPA) on the proliferation of 3AO,HHUA and DU145 cells in culture. Methods BPA(10^-9mol/L) or β-estradiol (β-E2, 10^-7 mol/L) was added to the culture of the three cell lines, and ethanol (1‰, V/V) was used as control group. 3AO(1 000 cells/well), HHUA(3 000 cells/well) and DU145(5 000 cells/well) were seeded on 96-well plates. Cell proliferation activity was examined by MTT colorimetric assays and the expression of Ki67 and PCNA were analyzed by semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR). The Elecsys System was used for immunoassay of CA15-3 in supernatant of HHUA cells culture. Results Compared with control group,the proliferation rates of 3AO,HHUA and DU145 cells in BPA group were 10.15%, 14.04% and 21.31% respectively,the effects of HHUA and DU145 cells with β-estradiol were 14.91%, 9.07%, but 13-estradiol had no effects on 3AO cell. Furt.hermore, the expression levels of Ki67 mRNA and PCNA mRNA of three cell lines in BPA group and HHUA and DU145 cells treated with β-estradiol were up-regulated significantly (P〈0.05); The content of CA15-3 in HHUA cells of BPA group was obviously higher compared with control group (P〈 0.05). Conclusion The findings in the present paper indicate that BPA can promote the proliferation of hormone-related tumor cells.
