电针对酒精中毒性周围神经病大鼠NGFmRNA表达的影响

摘　　要：目的：通过酒精自由饮建立慢性酒精中毒性大鼠周围神经病变模型,酶联免疫的手段和方法,从细胞分子生物学等不同层面探讨电针对慢性酒精中毒型周围神经病的治疗作用机制,发挥针灸治疗优势,为针灸治疗慢性酒精中毒性周围神经病的临床推广提供新的理论依据。方法：造模后的成年雄性Wistar大鼠50只,随机分成空白对照组、模型组、弥可保对照组、电针组、普通针刺组,每组各10只。各组按相应的方法进行治疗。结果：正常坐骨神经的NGFmRNA表达量极低,酒精中毒损伤后坐骨神经NGFmRNA表达增高,治疗2周达到一个小的高峰;2~4周NGFmRNA的表达出现一轻度回落趋势,但4周后又继续升高。针刺治疗后,治疗组的NGFmRNA表达较模型组均有不同程度升高,其中电针组升高幅度最为明显,始终处于高水平。结论：电针可升高酒精中毒性大鼠坐骨神经中NGFmRNA的含量。Objective：To discuss the mechanism of therapeutic effect on electro-acupuncture for treating the rat models of chronic alcoholism peripheral neuropathy on the level of cellular molecular biology.And to take advantage of acupuncture to offer theoretical basis for clinically developing the therapy.Methods：50 male adults＇ model WISTA rats with the weight of 200~250g were randomly divided into 5 groups：Control group,model group,Methycobal group,electro-acupuncture group,regular acupuncture group,10 for each group.The Methycobal group was injected Methycobal 300μg/kg/d into left and right lower limb muscles alternately for profitable absorption,and treatment was for 8 weeks.The electro-acupuncture group was needling the points of double Shenshu,Pishu,Housanli,Huantiao,Sanyinjiao,Taixi,connect Shenshu,Zusanli with slack-tight wave by KWD-808II pulse electronic treating machine for 20 minutes every 2 days for 8 weeks.Ordinary acupuncture group was treated the same way as electro-acupuncture group,but without electronic treating machine.Regular compare group and the model compare group were injected the same dosage of saline into muscle every day.Determine the expression of NGF in the rats＇ dorsal root ganglia and sciatic nerve tissue by the way of enzyme linked immunosorbent assay.Results：The expression of regular sciatic nerve＇s NGFmRNA is very low,while the one has increased after injury of chronic alcoholism,which reached one peak after 2 weeks treatment.2-4 weeks＇ treatment made the expression decrease for a while,but the quantity went up again after 4 weeks.Acupuncture made the treatment groups＇ expression of NGFmRNA increase with different degrees,the most obvious of which was the electro-acupuncture group.Conclusion：Electro-acupuncture can increase the content of NGFmRNA in the peripheral neuropathy of chronic alcoholism rats＇ sciatic nerve tissue.
