摘  要:目的:建立二阶导数光谱法测定溴吡斯的明聚乳酸微球的包封率。方法:通过离心法分离游离药物,采用二阶导数光谱法,于287nm波长处测定振幅D值,测定溴吡斯的明聚乳酸微球的包封率。结果:溴吡斯的明在16.68-38.92mg·L^-1范围呈良好线性关系;其回归方程为D=0.94×10^-2+0.66×10^-2(r=(1.9998);平均回收率100.30%、RSD为0.09%(n=3),24h内D值无明显变化,RSD为2.13%。结论:本方法操作简单、准确,可用于溴吡斯的明聚乳酸微球包封率的测定。OBJECTIVE To establish methods for the determination of the entrapment efficiency of polys(DL-lactide)microspheres containing pyridostigmine bromide. METHODS Centrifuging was applied to isolate free drug and second order derivative spectmphotometry was used. The peak amplitude was determined when the wavelength was at 287 nm. RESULTS The pyridostigmine bromide showed a good linear relationship within a range of 16. 68 - 38.92 mg· L^-1. The regression equation was D = 0. 94 × 10^-2 + 0. 66 × 10^-2 (r = 0. 9998) ; the average recovery was 100. 30% and RSD = 0. 09% (n = 3), numerical value D wasn't changed in the 24 h and RSD= 2.13%. CONCLUSION The method is simple, accurate and can be used for the determination of entrapment efficiency of poly(DL-lactide) mierospheres containing pyridostigmine bromide.
