化痰与活血基础方对非酒精性脂肪肝大鼠TNF-αI、L-6影响的实验研究

摘　　要：目的:研究NAFLD的发病机理,探讨化痰法与活血法在脂肪肝治疗中的作用效果,为临床治疗提供客观依据。方法:高脂饲料联合四氯化碳复制非酒精性脂肪肝(NAFLD)大鼠模型,选取化痰法的基础方二陈汤及活血法的基础方桃红四物汤对脂肪肝大鼠进行治疗,观察各组大鼠肝组织内肿瘤坏死因子-α(tumor necrosis factor,TNF-α)及白介素-6(interleukin-6I,L-6)的表达和含量。结果:正常组大鼠肝组织内TNF-α和IL-6含量极少,模型组大鼠肝组织中TNF-α和IL-6阳性表达面积和含量均有明显升高(P<0.01),免疫组化结果显示,二陈汤组与桃红四物汤组大鼠肝组织内TNF-α和IL-6阳性表达面积与模型组相比显著减少(P<0.01),对照组大鼠肝组织内IL-6阳性表达面积与模型组相比显著减少(P<0.01),TNF-α与模型组相比无统计学差异;抗体含量测定结果显示,二陈汤组大鼠肝组织内IL-6含量与模型组相比有所降低,差异有统计学意义(P<0.05);TNF-α与模型组相比无统计学差异;桃红四物汤组大鼠肝组织内IL-6与TNF-α与模型组相比均无差异。结论:二陈汤和桃红四物汤可以通过降低TNF-α和IL-6在肝组织内的表达起到抑制肝脏炎症反应和减少肝细胞损伤的作用,从而治疗脂肪肝,为化痰法和活血法治疗NAFLD提供理论依据。Aim：To explore the pathogenesis of Non-alcoholic fatty liver disease（NAFLD）,discuss how the phlegm dispersing method and promoting blood flow method take effect in the course of the treatment,in order to provide a basic for clinical intervention.Method：Experimental animal model of NAFLD were established,treated with Erchen decoction,Taohongsiwu decoction and compared with the control group and the model group.8 weeks later,we observed the distribution of the tumor necrosis factor-α（TNF-α） and inter leukin-6（IL-6）,and detected the content of them in the liver tissues by ELISA.Result：The level of TNF-αand IL-6 increased in the model group compared with the normal group（P0.01）.The distribution of TNF-αand IL-6 in the Erchen group and Taohongsiwu group was lower than them of the model group（P0.01）.The content of IL-6 in the Erchen group was lower than it of the model group（P0.05）.There was not different between the model group and the Taohongsiwu group The content of TNF-αof liver tissue in the Taohongsiwu group was not different from that in the model group.Conclusion：Erchen decoction and Taohongsiwu decoction can inhibit TNF-αand IL-6＇s expression to reduces inflammation and protect the liver cells.That provides an objective basis for the NAFLD＇s clinical treatment to use the phlegm dispersing method and promoting blood flow method.
