摘 要:目的探索兔骺板细胞和异种脱细胞软骨基质共同培养构建组织工程骺板的可行性。方法取2周龄新西兰白兔的骺板细胞,收取第二代细胞体外扩增后接种到脱细胞软骨基质上,混合培养,观察复合物组织相容性及病理情况。结果骺板细胞在脱细胞软骨基质上生长迅速,成活率高,未见明显的生长抑制;在体外培养可以形成组织块,分泌糖胺聚糖(Glycosaminoglycans,GAG)及Ⅱ型胶原等细胞外基质,但是细胞排列紊乱,没有典型的骺板组织结构,不能形成明显的骺板组织。结论异种脱细胞软骨基质和骺板软骨细胞在体外成功构建出组织工程骺板,为下一步研究奠定基础。Objective To study the construction of tissue-engineering growth plate with co-cultured epiphyseal plate cells and allograft articular cartilage extracellular matrix(CACEM).Methods Growth plate cells were collected from New Zealand rabbits at the age of 2 weeks,in vitro amplified,and inoculated into CACEM to observe their histocompatibility and pathological change.Results The growth plate cells grew rapidly on CACEM with a high survival rate and no significant growth inhibition.Formation of tissue mass,secretion of glycosaminoglycan(GAG),and type Ⅱ collagen could be observed when the cells were cultured in vitro.The cells were irregularly arranged with no formation of typical epiphyseal plate tissue.Conclusion Tissue engineering epiphyseal plate can be constructed with CACEM and growth plate cells in vitro,thus laying a foundation for further study.
