脯氨酸与2,3-二氯-5,6-二氰-1,4-对苯醌荷移反应的研究

摘  要:目的:采用分光光度法研究了脯氨酸与2,3-二氯-5,6-二氰-1,4-对苯醌(DDQ)的荷移反应。方法:在常温下,脯氨酸与DDQ在十二烷基硫酸钠溶液及pH=10.0的三酸缓冲溶液中,反应10 min,可生成稳定的荷移络合物。结果:荷移络合物的组成比为1∶1,最大吸收波长λmax=523nm,表观摩尔吸光系数ε=1.60×103L.mol-1.cm-1,在10μg ml-1~70μg ml-1的范围内符合比尔定律,回收率在97.7%~100.5%的范围内,桑德尔灵敏度S=7.2×10-2μg cm-2,相对标准偏差不大于1.5%。结论:应用本方法测定脯氨酸样品的含量,结果与文献方法一致。Objective:A simple,quick and sensitive spectrophotometric method has been established for the determination of proline.Methods: It was based on a charge transfer reaction of studied drug with 2,3-dichloro-5,6-dicyano-1,4-benzoquinone(DDQ) in Britton-Robinson(pH=10.0) buffer solution at room temperature for 10 min.Results: The composition of the complex of proline with DDQ was 1∶1.The apparent molar absorptivity of the complex was determined to be 1.6×103 L mol-1 cm-1 at 523 nm.The calibration curve was linear in the concentration range of 10 μg ml-1~70 μg ml-1 for proline.The relative standard deviation was less than 1.5% and the recovery was in the range of 97.7%~100.5%.Conclusion: This proposed method has been applied to the determination of proline in its pharmaceutical dosage forms with satisfactory results.
