摘 要:目的研究荞麦黄酮复方制剂(FBC)对2型糖尿病大鼠血糖、血脂和胰岛素抵抗(IR)的影响及机制。方法用高脂饲料和链脲佐菌素(STZ)建立大鼠2型糖尿病模型,随机分为模型对照组,阳性药物组(消渴丸0.83g/kg),FBC低剂量组(0.6 g/kg),FBC高剂量组(1.2 g/kg)和正常对照组,各组大鼠灌胃给药连续8周。用血糖仪测空腹血糖(FBG)、糖耐量(OGTT)、胰岛素敏感指数(ISI);用生化分析仪测定TG、TC、游离脂肪酸(FFA);放免法测定血浆空腹胰岛素(FINS);HE染色观察肝脏和胰腺组织病理学改变。结果与模型组比较,不同剂量的FBC能使FBG、TG、TC和FFA降低(P<0.05或P<0.01),ISI升高,并改善OGTT(P<0.05或P<0.01),能明显减轻肝和胰腺组织的病理改变。结论 FBC具有降血糖、改善IR的作用,其机制可能是通过调血脂作用产生。Objective To study the effect of flavones of buckwheat compound(FBC) on serum glucose,serum lipid and insulin resistance in type 2 diabetic rats and try to find its mechanism.Methods The models of diabetic type 2 rats with insulin resistance were made by injecting streptozotocin(STZ) and eating a high-fat diet.Rats were randomly divided into normal group,model group,FBC low and high(0.6 and 1.2 g/kg) groups and xiaoke group.Fasting blood glucose(FBG),OGTT,fasting insulin(FINS),insulin sensitivity dex(ISI),free fatty acids(FFA),triglyceride(TG),total cholesterol(TC) were measured after 8 weeks.The pancreas,liver tissues were stained by HE so as to observe its morphologic changes.Results FBG,TG,TC,FFA significantly reduced in the FBC groups as compared with those in the model group,while ISI significantly increased and improved OGTT(P〈0.05 or P〈0.01).Compared with the model group,the pathologic changes of the pancreas and liver tissues were significantly improved in different dose FBC groups.Conclusion FBC can significantly reduce FBG,improve insulin resistance by the mechanism of regulating serum lipid.
