摘 要:用随机引物扩增多态性DNA(RAPD)技术对白术的3个居群进行基因组DNA多态性分析,从100个随机引物中筛选出20个10bp随机引物,共扩增出187个DNA片段,片段大小在200bp~2800bp之间,其中多态性谱带为135条,表现出了丰富的RAPD多态性(占72.2%),根据遗传距离,利用UPGMA构建了居群亲缘关系柱状图。结果表明:於术86和於术76的亲缘较近,而於术86和白术天台之间的亲缘关系较远。就单个样本来看,於术86的15个样都聚在了一起,表明了於术86较强的同源性,於术76的14个样也聚到了一起,也表明了於术76的同源性。而於术76第14个样与白术天台聚到了一起,表明了於术76与白术天台有一定的同源关系。The fingerprints of 3 populations in genus Atractylodes were generated by the use of RAPD method. Twenty primers were screened from 100 random primers, and a total of 187 DNA fragments were amplified ranging from 200 bp to 2 800 hp, of which 135 bands were polymorphie (72.2%). The genetic distances were calculated and relationship dendrogram of A tractylodes was constructed by using UPGMA. The results indicated that the genetic relationship between YuZhu 86 and YuZhu 76 was close, and far between YuZhu 86 and Atractylodes Tiantai. Based on the cluster Analysis, the 15 samples from YuZhu 86 became a group, indicating a strong homology of YuZhu 86. However, 14 samples from YuZhu 76 gathered in a group. The 14th sample from YuZhu 76 clustered with A troctylodes Tiantai, It suggests that a homologous relationship existed between YuZhu 76 and Atractylodes Tiantai.
