摘 要:探讨了中华绒螯蟹亲蟹血清离子和渗透压对盐度的响应以及肝胰腺抗氧化能力与盐度的关系。设立4个盐度组(盐度6、12、18、24)和1个淡水对照组,检测第4天和第20天时中华绒螯蟹亲蟹血清中Na+、K+、Cl-浓度、渗透压以及肝胰腺超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性和还原型谷胱甘肽(GSH)、丙二醛(MDA)含量。结果显示,各实验组中华绒螯蟹亲蟹血清中离子和渗透压在实验过程中高于外界水体,处于高渗状态。各实验组间血清K+浓度无显著差异,Na+和Cl-浓度以及渗透压随着盐度梯度的升高而升高。实验第4天时,肝胰腺中CAT活性和MDA含量随盐度梯度的升高而显著降低(P〈0.05),其中CAT活性从0.479 1±0.089 5 U/mgprot降为0.052 3±0.012 1 U/mgprot;各实验组肝胰腺SOD活性和GSH含量无显著差异。至实验第20天时,各实验组肝胰腺SOD、CAT活性和GSH、MDA含量均未表现出显著性差异。实验结果表明中华绒螯蟹亲蟹在环境盐度0~24范围内进行高渗透压调节,盐度对中华绒螯蟹肝胰腺抗氧化系统具有一定影响。A trial with five different salinity concentrations was conducted to investigate the relationship between salinity and the osmoregulation and antioxidant capacity of hepatopancreas tissue in Parent Chinese mitten crabs(E.sinensis).The expreiments were settled with five different salinity concentrations,fresh water(control),salinity 6,12,18 and 24,three replications and 6 crabs per group.Chinese mitten crabs were transferred to the experiment tanks after 7 days of accommodation to the environment.Serum ionic density,osmotic pressure,the activity of superoxide dismutase(SOD),catalase(CAT) in hepatopancreas,content of reduced glutathione(GSH) and malondialdehyde(MDA) were monitored.Results showed that serum ionic density and osmotic pressure increased with the increasing of salinity concentrations when Chinese mitten crabs were cultured in lower osmolality condition.However,the increasing rate was lower than the change rate of rear water.No significant differences of K+ concentrations were observed among each group.The concentrations of Na+ and Cl-tended to increase with the increasing of salinity.The activity of CAT in hepatopancreas and the content of MDA decreased significantly with the increasing of salinity concentrations after four days.The activity of CAT in hepatopancreas decreased from 0.479 1±0.089 5 U/mgprot to 0.052 3±0.012 1 U/mgprot.There was no significant difference of the SOD activity in hepatopancreas and the GSH content among groups.No marked differences were observed in SOD,CAT of hepatopancreas and GSH,MDA content among groups after 20 days.It could be concluded that Parent Chinese mitten crabs was in the process of hyper osmoregulation when the rear salinity was 0-24.Antioxidant capacity of hepatopancreas in Chinese mitten crabs was affected slightly,and the response to the salinity was very short.
