摘  要:目的建立测定大鼠肝、肾组织中天麻素含量的方法。方法 Wistar大鼠分别灌胃天麻细粉混悬液后30min取出肝脏、肾脏,加0.9%生理盐水配置成匀浆液。用甲醇沉淀组织匀浆中的蛋白质,取上清液用氮气吹干,加双蒸水溶解后直接进样,用高效液相色谱仪测定。色谱柱:Phenomenence Luna C18(250 mm×4.6 mm,5μm);流动相:80%乙腈-0.1%磷酸(2.8:97.8);流速:1.0 ml.min-1;样品峰面积与内标检测波长:220 nm。天麻素保留时间(tR)约为15 min;柱温:30℃。结果肝、肾组织中天麻素的浓度在12.5~1 600μg.ml-1范围内,大鼠峰面积之比(Y=A/AIS)与天麻素的浓度X线性关系良好,回归后其标准曲线为Y=0.118 2X-2.174(r=0.999 7);定量限为12.5μg.ml-1;平均加样回收率为100.4%,RSD(5.0%。结论研究结果表明该方法简便、稳定、易行,可以用于测定大鼠肝、肾组织中天麻素的含量。Objective To establish a method for the determination of Gastrodin which is the main effective component of Tianma on rat liver and kidney.Methods The rat liver and kidney were removed after intragastric administration of Tianma suspension for 30 minutes.The samples were added normal 0.9% saline to obtain the homogenate solution.The tissue samples which were drawn and treated by adding methanol and the supernatant were dried by N2.After redissolving in water,the residues were detected by HPLC directly.The analysis was carried on an analytical column Phenomenence Luna C18(250 mm×4.6 mm,5μm).The mobile phase consisted of 80% acetonitrile-0.1% H3PO4(2.8:97.8) and the flow rate was 1.0 ml·min-1.The UV detection wavelength was at 220 nm and the column temperature was kept at 30℃.The retention time(tR) of Gastrodin was about 15 min.Results A good linearity was obtained from 12.5μg·ml-1 to 1600μg·ml-1 for gastrodin.Regression curve was y =0.1182x-2.174 with a correlation coefficient of 0.9997,the detection limit was 12.5μg·ml-1,the average recovery was 100.4%,and the RSD was less than 5.0%.Conclusion The method is simple,stable and easy for operation.It is shown to be suitable for determination of Gastrodin in rat liver and kidney.
