摘  要:目的研究道地产区的甘草经过栽培后遗传多样性的变化,从分子层面探讨中药"离其土,则失其味也"的科学性,为栽培甘草选育优良品种提供依据。方法采用ISSR-PCR技术对6组24个栽培甘草样品进行遗传多样性分析。结果用6条引物对24个样品DNA进行PCR扩增,共得到43个条带,其中29个条带具有多态性,多态性百分比为67.45%。用NTSYS-pc2.10分析软件进行聚类分析,基本可将不同道地产区的栽培甘草较好的区分,但同一产区种子栽培的甘草与种苗栽培的甘草并不能较好的聚类。结论栽培甘草不同产地遗传多样性都很丰富,同一产地的甘草由于不同的栽培代数遗传多样性有所改变,说明甘草经过多代有性繁殖后遗传多样性会有部分丢失,这可能与环境的选择有关,为优质甘草的栽培育种提供理论基础。Objective To detect genetic diversity of famous regions of cultivative Glycyrrhiza uralensis.and to discuss the scientificity of the famous remark " leaving the original environment,the medicines will miss their tastes "from the molecular level.Methods The genetic diversity of 6 groups,including 24 samples was investigated by ISSR maker technique.Results A total of 43 bands were produced in 24 samples,of which 29 bands were common in the 6 groups,which was about 67.45%.Groups cluster analysis based on the NTSYS-pc2.10 software indicated that most of the samples from different famous regions sources divided into different groups.But the samples from the same famous region and different cultivated generations could not be clustered together.Conclusion It shows genetic diversity of different source and genetic stability of the same source.The genetic diversity has changed because of the different cultivated generations.It shows that through many generations the genetic diversity of Glycyrrhiza uralensis will be partly lost.This may be related with the environmental choice,and the result can give a foundation of cultivating good quality Glycyrrhiza uralensis,seed selection and breeding of seeds.
