高效液相色谱法测定盐酸左旋多巴正丁酯的含量

摘　　要：目的建立测定盐酸左旋多巴正丁酯含量的高效液相色谱法。方法色谱柱采用WondailTM C18柱(150 mm×4.6 mm,5μm),流动相为0.05 mol/L磷酸二氢钠(用4.3%氢氧化钠溶液调pH至5.0)-甲醇(60:40),流速1.0 mL/min,检测波长280 nm,柱温40℃,进样量20μL。结果盐酸左旋多巴正丁酯质量浓度在25.1～251.0μg/mL范围内与峰面积线性关系良好(r=0.999 9),平均回收率为99.77%,RSD为0.83%(n=9)。结论高效液相色谱法简便、准确、专属性好,可用于盐酸左旋多巴正丁酯的含量测定。Objective To establish an HPLC method for the content determination of levodopa n-butyl ester hydrochloride.Methods The WondailTM C18 column（150 mm×4.6 mm,5 μm） was used and the mobile phase consisted of 0.05 mol/L sodium dihydrogen phosphate（adjusted to pH 5.0 with 4.3%sodium hydroxide solution）-methanol（60 ∶40） with the flow rate of 1.0 mL/min.The detection wavelength was 280 nm,the column temperature was 40 ℃and the injection volume was 20 μL.Results The linearity of levodopa n-butyl ester hydrochloride concentration was within 25.1-251.0 μg/mL（r=0.999 8）,the average recovery rate was 99.77%and RSD was 0.83%（n=9）.Conclusion This HPLC method is simple and accurate with good specificity,which can be applicable for the determination of levodopa n-butyl ester hydrochloride.
