波依定对压力负荷心肌肥厚大鼠心肌细胞凋亡相关基因c-myc表达的影响

摘　　要：目的探讨波依定(非洛平缓释片)对升主动脉缩窄压力超负荷心肌肥厚大鼠心肌细胞凋亡相关基因c-myc表达的影响。方法将升主动脉缩窄术后心肌肥厚Wistar雌性大鼠随机分为2组,心肌肥厚组,给予生理盐水,1次/d灌胃(n=10);治疗组,给予波依定,1次/d灌胃(n=10);治疗4周。同时制备模型设置假手术组作为对照(n=10),不予处置。观察各组大鼠各项指标变化。结果心肌肥厚鼠心肌细胞c-myc蛋白表达显著增高,波依定药物治疗4周后,心肌细胞内c-myc蛋白表达降低。结论心肌细胞内c-myc蛋白表达与心肌肥厚密切相关,波依定可降低肥厚心肌内c-myc蛋白表达,缓解心肌肥厚症状。Objective To study the effects of Plendil on the expression of c-myc of cardiac myocyte of cardiomyopathy pressure overload rat models subsequent to coarctation of ascending aorta. Methods A model of cardiomyopathy induced by coarctation of ascending aorta was used in this study. 20 female Wistar cardiomyopathy rats were divided randomly into two groups （n=10）. The control group received saline butfer （once a day）.The treatment group received Plendil （once a day）.Treatment time was 4 weeks. Ratio of LV weight to body weight, expression of c-myc were investigated in the two groups and sham operation group.Results c-myc expression was higher in cardiomyopathy when compared with control. Plendil could decrease the expression of c-myc level.Conclusion These result indicated that cardiomyopathy was probably related to activating of c-myc. Plendil may contribute to the observed beneficial effects on cardiomyopathy by the decrease of c-myc expression.
