摘 要:为探讨大鼠尿液游离皮质酮昼夜节律性变化,采集24 h内8个时间段大鼠尿液,应用ELISA法检测了尿游离皮质酮含量,生化法检测了尿肌酐含量,以皮质酮与尿肌酐比值反映尿游离皮质酮水平,对各个时间段尿游离皮质酮水平的生物时间序列进行了宏观分析与比较。结果表明,大鼠尿液游离皮质酮与尿肌酐比值呈现昼夜波动性;尿液游离皮质酮的检测作为一种非侵入性检测方法,采样方便,对动物无损伤,避免了采血过程对动物皮质酮激素的干扰,有利于长期动态观察大鼠皮质酮激素,增强实验的可靠性。To explore the circadian rhythm of urine dissociative corticosterone in rats,the rats urine samples were collected in eight periods of 24 h.The concentrations of urine dissociative corticosterone and creatinine were detected respectively by enzyme-linked immunosorbent assay(ELISA) and biochemical method.All biological time-series of urine dissociative corticosterone level were analyzed with macroscopic analysis.The results showed that the ratio of urine dissociative corticosterone and creatinine showed diurnal fluctuation.The detection of urine dissociative corticosterone is non-invasive,and convenience sampling,has no damage to animals,avoid the interference from blood sampling process on corticosterone hormone,so it helps to long-term dynamic observation of rat corticosterone hormone,and enhances the reliability of experiment.
