摘 要:目的:研究疼痛对大鼠血清单胺类递质含量的影响。方法:大鼠左后足跖底部皮下注射5%甲醛建立动物疼痛模型,进行行为学观察,采用荧光分光光度法测定正常和模型大鼠血清中去甲肾上腺素(NE)和多巴胺的含量,NE的激发/发射波长分别为384/483 nm,多巴胺的激发/发射波长分别为324/373 nm。结果:NE线性范围是:0.01~0.25μg/ml(r=0.996 5,n=7);多巴胺线性范围是:0.01~0.20μg/ml(r=0.993 5,n=7)。NE和多巴胺的日内、日间RSD均〈20%,相对回收率分别为99.8%~113.5%和99.4%~106%。正常组大鼠血清NE和多巴胺含量分别为(0.088±0.030)(、0.047±0.005)μg/ml,模型组则分别为(0.122±0.010)、(0.064±0.010)μg/ml。结论:荧光分光光度法测定大鼠血清中NE、多巴胺含量准确、灵敏。皮下注射5%甲醛可致疼痛症状产生,疼痛可能引起大鼠血清中NE和多巴胺水平升高。Objective: To investigate the effect of pain on contents of monoamine neurotransmitters in serum of pain model rats.Methods: The model was established by subcutaneous injection of 5% formaldehyde at the sole of left hind limb and the animal behavior was observed and recorded.Contents of norepinephrine(NE) and dopamine in serum of normal and model rats were determined by fluorescence spectrophotometry.The excitation emission wavelength of NE was 384 nm/483 nm,and that of dopamine was 324/373 nm.Results: The linear response of NE ranged from 0.01-0.25 μg/ml(r=0.996 5,n=7),that of dopamine ranged from 0.01-0.20 μg/ml(r=0.993 5,n=7).Intra-day and inter-day RSD of both NE and dopamine were less than 20%.The relative recovery rates of NE and dopamine were 99.8%-113.5% and 99.4%-106.0%,respectively.The contents of NE and dopamine in serum of rats in control group were(0.088±0.030),(0.047±0.005) μg/ml,and those of the rats in model group were(0.122±0.010),(0.064±0.010) μg/ml,respectively.Conclusion: Fluorescence spectrophotometry is accurate and sensitive for the determination of NE and dopamine in serum of rats.Subcutaneous injection of formaldehyde engenders symptoms of pain and probably raises the contents of NE and dopamine in serum of pain model rats.
