摘 要:目的:优选丹黄凝胶方中有效成分的最佳提取工艺。方法:采用HPLC法测定指标成分大黄酚、小檗碱的含量,以大黄酚、小檗碱的提取率及干膏得率为工艺考察指标,按U8(82×42)均匀设计拟水平表安排实验,分别考察了乙醇浓度、乙醇用量、提取次数和提取时间4个因素的影响。结果:通过DPS统计软件分析,得出丹黄凝胶方的最佳提取工艺为7倍量的65%乙醇,提取3次,每次2 h。在此提取工艺下大黄酚、小檗碱的提取率分别为85.75%和85.11%,干膏得率为36.12%。结论:均匀设计法优选出的丹黄凝胶方提取工艺稳定可行,为丹黄凝胶方的进一步研究提供了参考。Objective:To optimize the best conditions for extraction of the active components in Danhuang gel.Methods:HPLC method was used to determine the concentrations of the index components chrysophanol and berberine.The extracting process of Danhuang gel was optimized by U8(82×42)uniform design with the extract rates of chrysophanol and berberine,dried extract weight as the indexes to observe the effects of ethanol concentration,amount of ethanol solvent added,frequency of decoction and duration of decoctions.Results:Through the DPS software analysis,the most favorable conditions were as follows:the active components in Danhuang gel were extracted in 65% ethanol 7 times as much as the gel for 3 times and 2 h every time.Under the above conditions,the extract rates of chrysophanol and berberine were 85.75% and 85.11%,respectively.The dried extract weight was 36.12%.Conclusion:The extraction technique for the active components-chrysophanol and berberine in Danhuang gel by uniform design is stable and feasible.The study could provide the references for determination and extraction of the active components in Danhuang gel and the information for its further investigation.
