蓬草除痹汤对Ⅱ型胶原蛋白诱导性关节炎大鼠血清IL-1β、TNF-α及PGE_2影响的实验研究

摘　　要：目的:观察蓬草除痹汤对Ⅱ型胶原蛋白诱导关节炎(CIA)大鼠血清IL-1β、TNF-α及PGE2的影响及作用机制。方法:建立Ⅱ型胶原蛋白诱导大鼠类风湿性关节炎模型,随机分为蓬草除痹汤高、中、低剂量组、雷公藤组及空白组,观察6周,评价蓬草除痹汤对CIA大鼠右足踝关节肿胀度的抑制作用,对体重及外周血清IL-1β、TNF-α及PGE2水平的影响。结果:与模型组比较,各用药组可显著抑制大鼠右足踝关节肿胀的程度(P<0.01,P<0.05);蓬草除痹汤中、高剂量组均可明显提高大鼠体重增加速度(P<0.01,P<0.05);各用药组改善血清IL-1β、TNF-α及PGE2水平均明显优于模型组(P<0.01),其中蓬草除痹汤中、高剂量组的抑制作用明显优于雷公藤多甙片组(P<0.05)。结论:蓬草除痹汤能有效改善CIA大鼠关节炎症状,其机制可能与下调血清中致炎因子IL-1β、TNF-α及PGE2的水平有关,值得进一步开发研究。Objective ： To observe the effect and mechanism of Pengcao Chubi Decoction on serum IL - 1β, TNF - α and PGE2 in rats with arthritis induced by Type Ⅱ collagen. Methods： The model rats with rheumatoid arthritis induced by Type Ⅱ collagen were randomly divided into high, medium and low dose group of Pencao Chub! Decoction, tripterygium group and control group. Six weeks later evaluated the inhibition of Pengcao Chubi Decoction on CIA rats joint swelling of the right foot and its effect on the body weight and the levels of peripheral blood IL - 1β, TNF - α and PGE2. Results： Compared with model group, treatment groups can significantly inhibit the degree of swelling of the right foot （ P 〈 0. 01, P 〈 0. 05 ） ; the rate of weight gain can be sig- nificantly improved （ P 〈0.01, P 〈0.05 ） in the high dose group and medium dose group of Pencao Chubi Decoction; the im- provement of the serum IL - 1 β, TNF - α and PGE2 in the treatment groups was significantly better than that of the model group （P 〈0.01 ）, in which the inhibitory effect of high dose group of Pencao Chubi Decoction was significantly better than that of tripterygium group （ P 〈 0. 05 ）. Conclusion ： Pengcao Chubi Decoction can improve arthritic symptoms, which may be associated with its function in regulating the levels of serum IL - 1 β, TNF - α and PGE2, which is worthy of further research and development.
