2型糖尿病大鼠大脑皮质HO-1表达的变化

摘  要:目的:探讨2型糖尿病大鼠大脑皮质血红素加氧酶1(HO-1)表达的变化。方法:20只SD大鼠随机分为正常对照组和糖尿病模型组,每组10只。正常对照组大鼠不做任何处理;糖尿病模型组大鼠采用链脲佐菌素连续腹腔注射建立2型糖尿病大鼠模型,一周后以血糖≥16.7mmol/L作为成模标准。HE染色观察大脑皮质神经细胞的变化,分别采用WesternBlotting法和RT-PCR法检测大脑皮质HO-1蛋白及mRNA的表达。结果:糖尿病模型组大鼠大脑皮质神经元细胞核浓缩,结构不清,出现神经元坏死迹象,糖尿病模型大鼠大脑皮质HO-1蛋白及mRNA的表达均明显高于正常对照组大鼠(P<0.01)。结论:2型糖尿病大鼠大脑皮质HO-1的表达明显上调,可能是糖尿病神经系统损伤的机制之一。Objective:To investigate the changes of hemeoxygenase-1 (HO-1) expression in cerebral cortex of type 2 diabetic rats.Methods:20 male SD rats were randomly divided into normal control group and diabetes model group(n=10). The diabetes model rats were made by eontinuous intraperitoneal injection of streptozotocin and blood glucoses〉 16.7mmol/L was taken as the standard after 1 week. The morphological changes of neurocytes were observed by HE staining. Western Blotting was used to detect the expression of HO-1 protein and RT-PCR was used to detect the expression of HO-1 mRNA in cerebral cortex.Results:Degeneration of neurocytes and inspissation of karyon were observed in cerebral cortex of diabetic rats. The expression of HO-1 protein and mRNA in cerebral cortex of diabetic rats were obviously higher than normal control rats(P〈0.01).Conclusions:The HO-1 expression in cerebral cortex of diabetic rats is obviously up regulated, this maybe one of the mechanisms of nervous system injury during diabetes mellitus.
