大豆苷元对HEC-1B细胞增殖及雌激素受体报告基因表达的影响

摘　　要：目的探讨大豆苷元对子宫内膜癌细胞增殖的影响及其对雌激素受体报告基因表达的影响。方法体外培养子宫内膜癌细胞子宫内膜癌雌、孕激素受体阴性细胞系(HEC-1B),采用四甲基偶氮唑盐法(MTT)检测不同浓度大豆苷元对子宫内膜癌细胞增殖的影响,同时检测大豆苷元对雌激素应答原件(ERE)调控的雌激素受体α(ERα)和雌激素受体β(ERβ)的荧光素酶报告基因表达的影响。结果大豆苷元对HEC-1B细胞体外增殖有明显抑制作用,差异有统计学意义(P<0.01),其抑制作用有明显量效和时效性特征。在浓度为20～80×10-6mol/L的大豆苷元作用下,报告基因luc的表达较正常对照组明显提高,差异有统计学意义(P<0.05),在80×10-6mol/L时到达最高值;大豆苷元通过ERβ介导的报告基因表达水平的升高程度强于ERα,差异有统计学意义(P<0.01)。结论大豆苷元可通过调节子宫内膜癌细胞ERα和ERβ介导的报告基因的表达,调整ERα/ERβ比例,从而抑制子宫内膜癌细胞的增殖。Objective To study the effect of daidzein on the proliferation of endometrial carcinoma in uterus and its mechanism.Methods Endometrial carcinoma cell HEC-1B were cultured in vitro,the influences of genintein in different doses on the proliferation of HEC-1B cell were detected by MTT method,and the expression of luciferase reportor gene of ERα and ERβ effected by ERE were evaluated.Results Daidzein inhibited significantly the proliferation of HEC-1B cells in vitro（P0.01） and the effect was streghtened along with the increase of the dose and the prolong of action time.Under the control of daidzein concentration 20～80×10-6mol/L,the expression of reporter gene luc was increased compared with control group（P0.05） and reached the highest point in the concentration 80×10-6mol/L;the increase of reporter gene expression induced by ERβ was stronger than that induced luc expression either through ERα（P0.01）.Conclusion Daidzein can inhabit the proliferation endometrial carcinoma through regulating the expression of ERα or ERβ,and regulating the balance of ERα/ERβ.
