摘 要:目的探讨IκB分子的上游蛋白激酶(IKKβ)在幽门螺杆菌相关性胃炎(HAG)病理变化的可能作用机制及灭幽汤的影响效应。方法建立HAG模型,将造模成功的小鼠按体质量随机分为模型组、丽珠得乐组,灭幽汤组。采用免疫组化法检测胃黏膜组织中核转录因子(NF-κB)、IKKβ的蛋白表达情况。HE染色观察胃组织病理变化。结果灭幽汤组能明显减轻胃组织的炎症反应,灭幽汤组胃黏膜炎症程度低于模型组,差异有统计学意义(P<0.05);能抑制HAG胃组织中NF-κB、IKKβ蛋白的活性,灭幽汤组NF-κB、IKKβ表达强度明显低于模型对照组,差异有统计学意义(P<0.05)。结论 NF-κB和IKKβ在HAG的炎症反应中发挥重要的作用,灭幽汤治疗HAG的可能作用机制是通过抑制IKKβ的激活,阻止其活化。Objective To discuss the effects of Mieyou decoction on IKKβ in Helicobacter pylori-associated gastritis(HAG) and its possible mechanisms on pathological changes.Methods After HAG models were established,the mice were randomly divided into model,Cabernet Franc Sidel and Mieyou decoction groups according to the weight.The expression of NF-κB and IKKβ protein in gastric mucosa tissue were detected with immunohistochemical and gastric tissue pathological changes with HE staining.Results The mucosal inflammation in Mieyou decoction group was significantly lighter than that in model control group(P0.05);inhibit the activities of NF-κB and IKKβ,the expression intensity of NF-κB and IKKβ was lower than that in model group(P0.05).Conclusion NF-κB and IKKβ play an important role in HAG inflammation,and the possible mechanism of Mieyou decoction is to inhibit activation of IKKβ to prevent activation of NF-κB.
