摘  要:目的对远志药材水解产物进行分离,鉴定,初步探讨其水解规律。方法远志药材用稀醇提取,正丁醇萃取,大孔吸附树脂富集得到总皂苷,总皂苷酸水解后,用硅胶色谱柱、制备液相方法分离得单体化合物,利用紫外光谱、核磁共振、质谱法等鉴定技术,确定单体化合物结构。结果从远志皂苷水解产物中分离得到了4个单体化合物,分别为,远志酸(1),远志皂苷元(2),远志酸-3-O-β-D-葡萄糖苷(3),远志皂苷元-3-O-β-D-葡萄糖苷(4),其中3、4为新化合物。结论发现了一条成熟的高效制备远志酸对照品的路线,为进一步完善远志质量标准奠定了基础。Objective To isolate and identify the hydrolysis product of radix polygalae, to elucidate rule of hydrolization initially. Methods Polygalae was extracted by diluted alcohol, was enriched by nbutanol and macroporous resin. The total saponin was hydrolyzed by hydrochloric acid. The separation was performed on silica gel column by high performance liquid chromatography (HPLC). The structure of compounds were identified through ultraviolet (UV), nuclear magnetic resonance ( NMR), mass spectrometry (MS). Results Four compounds from hydrolysis product of Radix polygalae were identified, polygalic acid ( 1 ), senegenin (2), polygalic acid - 3 - 0 - β - D - glucoside ( 3 ) and senegenin- 3 -O-β- D- glucoside (4). Conclusion This method is efficient to obtain polygalacic acid and provide a basis to improve the quality standard of radix polygalae further.
