摘  要:从东祁连山高寒草地采集优势牧草紫花针茅并分离其内生细菌,以辣椒立枯丝核病菌、油菜菌核病菌、番茄早疫病菌、番茄灰霉病菌、玉米大斑病菌、小麦离蠕孢、甜瓜枯萎病菌和玉米小斑病菌8种植物病原真菌为指示菌,筛选拮抗菌,用阿须贝无氮培养基和PKO培养基筛选固氮菌和溶磷菌,采用传统生理生化测定法和16S rDNA基因序列同源性分析对筛选出的内生细菌进行鉴定。结果从紫花针茅分离到6株内生细菌,其中,菌株ZH4和ZH6对辣椒立枯丝核病菌生长有抑制作用,平板对峙时的抑菌带宽分别为5 mm和4 mm;没有分离到固氮菌和溶磷菌。ZH4和ZH6菌体均为杆状,革兰氏阳性,产芽孢,16S rDNA序列在GenBank中登录号分别为EU236750和EU236752,结合生理生化特征及16S rDNA基因序列同源性分析,鉴定ZH4和ZH6均为蜡状芽孢杆菌。Endophytic bacteria were isolated and screened from dominant grass Stipa purpurea on alpine grassland in the Eastern Qilian Mountains,and their characteristics of anti-pathogenic fungi,nitrogen-fixing and Phosphate-solubilizing were tested by using eight plant pathogenic fungi Ashby Nitrogen-free medium and Pikovaskaia's medium methods,identified bsaed on their.Physiological and biochemical characteristics and 16S rDNA gene sequence analysis.The rerults showed that six endophytic bacteria strains were isolated from Stipa purpurea.Strains ZH4 and ZH6 had an inhibitive effect on the growth of Rhizoctonia solani kuhn and inhibitory radius was 5 mm and 4 mm in Plate confrontation experiment respectively.There were no nitrogen-fixing bacteria and phosphate-solubilizing bacteria isolated.Strains ZH4 and ZH6 were Rod-shaped,Gram-positive,and produce spore.16S rDNA sequence of ZH4 and ZH6 have got Gene Bank accession numbers EU236750 and EU236752,and was identified as Bacillus cereus.
