摘 要:目的探讨急性损伤对大鼠脊髓血管内皮生长因子(VEGF)两个受体(Flt-1、Flk-1)表达的影响。方法采用改良Allen's重量打击法大鼠急性脊髓损伤模型,用免疫组织化学方法观察急性损伤后大鼠脊髓VEGF受体Flt-1、Flk-1表达的动态变化。结果 Flt-1、Flk-1在大鼠脊髓损伤后12 h表达上调,24h达高峰,7d时开始下降。表达细胞主要为神经胶质细胞,其次为血管内皮细胞和巨噬细胞。Flk-1表达细胞比Flt-1更多见。结论急性损伤可上调Flt-1、Flk-1在脊髓血管内皮细胞、胶质细胞和巨噬细胞内的表达,VEGF受体Flt-1、Flk-1可能通过保护血管内皮细胞和促进血管内皮细胞生长,对损伤神经组织起到一定的保护作用。Objective To investigate the time course of vascular endothelial growth factor receptor (Flt-1, Flk-1) expression in rats spinal cord following acute injury. Methods The spinal cord injury model was made by modulated Allen's weight drop method. The expressions of Flt-1, Flk-1 were assayed by using immunohistochemical staining. Results Flt-1, Flk-1 expressions were up-regulated at 12 hours after the injury, and they reached the peak at 24 hours. On 7 days after the injury, the expressions decreased. The expressions were mainly in astrocytes, and next in vascular endothelial cells and macrophages. The positive cells of Flk-1 were more than those of Flt-1. Conclusion Acute injury can induce the expressions of vascular endothelial growth factor receptor. Vascular endothelial growth factor and vascular endothelial growth factor receptor may play an important role in acute spinal cord injury by the maintenance and induction of growth of vascular endothelial cells.
