摘  要:采用实时荧光定量RT-PCR法,分别于4、8、12、16、20和24 h时,检测了不同浓度苦参碱对小菜蛾幼虫体内γ-GABA受体基因的相对表达量。结果表明,在处理4 h时,以为15.625×10-3mg/mL的苦参碱处理小菜蛾,其γ-GABA受体基因的表达量达到最大值,为23.62,在20 h时,γ-GABA受体基因的表达量达到最小值,为11.456。在4、8、12、16和20 h时,125×10-3mg/mL的苦参碱处理小菜蛾幼虫的γ-GABA受体基因的表达量均低于其他的处理,在24 h时,同样浓度处理小菜蛾,其γ-GABA受体基因的表达量高于其他处理,差异显著,在16 h时,同样的浓度处理小菜蛾幼虫γ-GABA受体基因的表达量低于其他处理,达到最小值,为4.345。由此说明苦参碱抑制了小菜蛾体内γ-GABA受体基因的表达。Effect of matrine on γ-aminobutyric acid receptor gene expression of diamondback moth,Plutella xylostella(L.) were investigated.The relative γ-aminobutyric acid receptor gene expression of P.xylostella larvae treated with matrine at 4,8,12,16,20,and 24 h,respectively,was analyzed by real-time quantitative RT-PCR.The results showed that the relative γ-GABA receptor gene expression of P.xylostella larvae treated with 15.625 × 10^-3 mg/mL of matrine reaches its maximum value of 23.620 and a minimum value of 11.456 at 4 h and 20 h,respectively.The relative γ-GABA receptor gene expression of P.xylostella larvae treated with 125×10^-3 mg/mL of matrine was lower than other concentrations at 4,8,12,16 and 20 h,respectively,while the relative γ-GABA receptor gene expression of them at 24 h was higher than other concentrations significantly.The relative γ-GABA receptor gene expression of them at 16 h reached a minimum value of 4.345 which was lower than the other treated concentrations of matrine.It also indicated that the relative γ-GABA receptor gene expression of diamondback moth larvae was inhibited by matrine.
