摘  要:金电极在含有1.0 mol/L KCl的0.1 mol/L吡咯溶液中,-0.3~0.8 V循环扫描10圈制备了聚吡咯膜,在1.2 V进行过氧化处理,得到聚吡咯微孔膜,并将双链小牛胸腺DNA沉积在膜的微孔内,利用差分脉冲技术以亚甲基蓝(MB)为电化学指示剂研究了新型有机污染物全氟辛烷磺酸(PFOS)对DNA的损伤。结果表明随着损伤时间的增加,MB还原峰电流减小,表明PFOS与DNA的相互作用阻碍了碱基鸟嘌呤G与MB的结合;同时电极的表面电子传递电阻从650 W增大到1350 W,表明PFOS对DNA的损伤降低了DNA的电子传递性能。该DNA传感器为研究污染物的毒理提供了新思路。Thepolymerization of pyrrole was fabricated on gold electrode by cyclic scan in the solution of 0.1 mol/L pyrrole containing 0.1 mol/L KCl between-0.3 and 0.8 V at a scan rate of 50 mV/s for 10 cycles.Then,the PPy membrane was electrochemical oxidated of at +1.2 V.The film is porosity.Following,the double-stranded calf thymus DNA was deposited within the pores.DNA damage induced by organic pollutant PFOS was studied by the differential pulse voltammetry(DPV) and take methylene blue(MB) as electrochemical indicator.The results show MB reduction peak current decreases with the incubation time increasing and the ionic strength enhanced.It indicates that the interaction of PFOS and the DNA hinders the combination of guanine(G) and MB.The electronic transfer resistance has increased from 650 W to 1350 W.It indicated DNA damage due to PFOS reduces the electron transfer properties.The DNA biosensor provides a novel avenue to research the pollutant toxicology.
