喘可治注射液增强细胞因子诱导的杀伤细胞抗肿瘤活性的初步研究

摘　　要：目的通过喘可治注射液对细胞因子诱导的杀伤细胞(CIK)的表型及细胞因子分泌情况的影响,探讨喘可治注射液对CIK免疫活性的免疫调节作用。方法分离健康志愿者的外周血单个核细胞,在细胞因子的作用下,体外培养成细胞因子诱导的杀伤细胞(CIK),在培养过程中同时加入喘可治注射液。LDH法检测不同浓度喘可治注射液培养条件下的CIK细胞的抗肿瘤活性。流式细胞术检测CIK细胞表型及细胞内因子的分泌情况变化。Annexin V/PI凋亡试剂盒检测CIK细胞的凋亡情况。结果喘可治注射液能提高CD3+CD56+双阳性的CIK细胞的比例,增强其杀伤肿瘤细胞的活性,促进具有抗肿瘤效应的Th1型的细胞内因子IFN-γ、TNF-α的分泌,而对CD4+CD25+抑制性细胞和Th2型的细胞因子IL-4的分泌影响不大。喘可治注射液对CIK细胞的凋亡没有明显作用。结论喘可治注射液可上调CIK细胞分泌Th1型的细胞因子,增强CIK细胞的抗肿瘤活性。Objective To explore the immune regulation of chuankezhi injection on cytokine-induced killer cell（CIK）through its impacts on phenotype and cytokine secretion of CIK.Methods The peripheral monocytes were isolated from healthy donors.Under the action of cytokine,the isolated cells were cultured as CIK in vitro.During the culture,chuankezhi injection was added.LDH assay was used to detect the anti-tumor activity of CIK cultured with chuankezhi injection under different concentrations.Flow cytometry was applied for detecting the changes in CIK phenotype and cytokine secretion.AnnexinⅤ/PI apoptosis kit was used to detect CIK apoptosis.Results Chuankezhi injection increased the proportion of CD3＋CD56＋ double-positive CIK,enhanced the activity of killing cancer cell and promoted the secretion of Th1 cytokines,IFN-γ and TNF-α that acted on anti-tumor.But,the injection had not a big impact on the secretion of CD4＋CD25＋ suppressor cells and Th2 cytokine,IL-4.Additionally,the injection had no apparent action on the apoptosis of CIK.Conclusion Chuankezhi injection up-regulates CIK for Th1 cytokine secretion and enhances the anti-tumor activity of CIK.
