摘  要:为改善胆红素氧化酶性质,提高酶的使用效率,以海藻酸钠为载体,采用包埋法固定了胆红素氧化酶,并优化了固定化条件,研究了固定化胆红素氧化酶的酶学性质。结果表明:海藻酸钠的质量浓度为0.10 g·mL-1、CaCl2浓度为0.20 mol·L-1、固定时间为1 h、酶的固定化效果最好,固定化酶的平均酶活力回收率达32%,固定化酶的最适催化pH值为7.3,相比游离酶降低了0.8;固定后酶的耐热温度较游离状态时提高15℃,说明热稳定性得到提高;固定化酶重复使用8次后仍保留原固定化酶活力的50%,使用稳定性也较高。An experiment was conducted to study the property of bilirubin oxidase(BOD) using sodium alginate as a carrier in order to improve properties of BOD and enhance efficiency of the enzyme.The immobilization condition of the BOD was optimized.The optimal conditions for BOD were obtained as sodium alginate 0.10g·mL^-1,CaCl2 concentration 0.20mol·L^-1,and immobilization time 1h.The average recovery rate of immobilized BOD activity was 32% and the optimum pH for the immobilized BOD activity was 7.3 under the optimal conditions,which reduced 0.8 compared with that of free enzyme.Heat stability of the immobilized enzyme was raised by 15 degrees C.The immobilized enzyme maintained its activities at a high level(50 percent) after being used eight times,indicating that the immobilized enzyme has good heat stability and could be used repetitiously.
