摘 要:目的观察正己烷对大鼠卵巢颗粒细胞凋亡调控基因bcl-2、bax、bcl-xl、xiap mRNA表达的影响。方法 48只成年雌性Wistar大鼠按体重随机分为4组,分别给予浓度为0,3.01,9.03,27.1 g/m3正己烷静式吸入染毒;每天4 h,每周6 d,持续6周;染毒结束待大鼠进入动情期,处死并提取卵巢颗粒细胞,并采用实时荧光定量PCR法测定颗粒细胞bcl-2、bax、bcl-xl、xiap mRNA表达水平。结果随着染毒剂量增加,各组bcl-2 mRNA表达呈下降趋势(2-△△CT值分别为1.069,0.762,0.698,0.690),bax mRNA表达呈上升趋势(2-△△CT值分别为1.039,1.242,1.317,1.895),各染毒组与对照组比较,差异均有统计学意义(P<0.05)。各染毒组bcl-xl mRNA表达与对照组差异无统计学意义(P>0.05);27.1g/m3染毒组xiap mRNA表达(0.694)低于对照组(0.999),差异有统计学意义(P<0.05)。结论正己烷上调大鼠卵巢颗粒细胞bax,下调bcl-2、xiap mRNA表达,可能是正己烷致使卵巢颗粒细胞凋亡的机制之一。Objective To observe the effects of n-hexane on the expression of apoptosis regulator gene bcl-2,bax,bcl-xl,and xiap mRNA in rat ovarian granulosa cells.Methods Totally 48 adult female rats were randomly assigned to four groups based on weight and were exposed to 0,3.01,9.03,and 27.1 g/m3 n-hexane for 6 weeks(4h/d,6 days/week),respectively.After n-hexane inhalation,the rats in estrus stage were killed and their ovarian granulosa cells were collected.The expressions of bcl-2、bax,bcl-xl,and xiap mRNA were measured with real-time fluorescence quantitative PCR method.Results With the increased n-hexane concentration,the expression of bcl-2 mRNA decreased(the value of 2-△△CTwas 1.069,0.762,0.698,0.690,respectively)and the expression of bax mRNA increased gradually(the value of 2-△△CTwas 1.039,1.242,1.317,1.895,respectively),both of them in n-hexane groups showed significant difference compared with those of in the control group(P0.05).The bcl-xl mRNA expression in all n-hexane groups showed no significant difference compared with that of the control group(P0.05).The expression of xiap mRNA in 27.1g/m3 group(the value of 2-△△CTwas 0.694) decreased significantly compared with that of in the control group(the value of 2-△△CTwas 0.999,P0.05).Conclusion N-hexane could enhance bax mRNA expression and downregulate bcl-2 and xiap mRNA expression,which might be the mechanism of apoptosis of rat ovarian granulosa cell.
