摘 要:目的观察白蒺藜有效组分对自发性高血压大鼠(SHR)心肌纤维化以及相关细胞因子表达的影响,探讨白蒺藜有效组分改善心室重构、抑制高血压靶器官损伤的可能作用机制。方法 32只14周龄雄性自发性高血压大鼠随机分为模型组、卡托普利组、白蒺藜组、联用组,每组8只,同周龄雄性Wistar大鼠8只作为正常对照。卡托普利组采用卡托普利30 mg/(kg.d)、白蒺藜组采用白蒺藜有效组分15 mg/(kg.d)、联用组采用白蒺藜有效组分15 mg/(kg.d)同时给予卡托普利30 mg/(kg.d)、模型组及正常组给生理盐水2 mL/(kg.d)灌胃,共8周。分别采用HE及VG染色法观察心肌病理变化及纤维化程度;ELISA法检测心肌转化生长因子β(TGFβ1)及结缔组织状因子(CTGF)表达变化。结果与Wistar大鼠比较,14周龄SHR大鼠收缩压明显升高(P<0.01);8周后,模型组大鼠收缩压较14周龄时明显升高(P<0.05)。与模型组相比,白蒺藜组收缩压明显下降(P<0.01)。VG病理染色心肌病理形态结果提示,22周龄SHR出现心肌纤维化改变,各用药组均能改善心纤维化程度,且改善程度相当。SHR大鼠心肌TGFβ1及CTGF较正常组表达显著增加(P<0.05)。与SHR相比,卡托普利能明显下调SHR心肌TGFβ1表达(P<0.05);而白蒺藜有效组分可显著下调TGFβ1及CTGF水平(P<0.05)。卡托普利及白蒺藜联用可明显下调TGFβ1(P<0.01)及CTGF水平(P<0.05)。结论白蒺藜有效组分能降低SHR收缩压,并从形态学角度证实其具有抑制心肌纤维化作用。自发性高血压大鼠心室肌TGFβ1及CTGF表达明显增加,白蒺藜有效组分可抑制这一作用,从而明显改善高血压所致心脏结构重塑。Objective To observe the influences of the effective components of Baijili(Fructus Tribuli) on myocardial fibrosis and expressions of relative cytokines in spontaneously hypertension rats(SHR),and discuss the possible mechanism of the effective components of Baijili in ameliorating ventricular remodeling and inhibiting the lesion of hypertensive target organs.Methods Male SHR(n=32,14 weeks old) were randomly divided into the model group,captopril group,Baijili group and combination group(each n=8),and other Wistar rats(n=8,same aged) were included into the normal group.The captopril group was given captopril(30 mg/kg/d),Baijili group was given the effective components of Baijili(15 mg/kg/d),combination group was given the effective components of Baijili(15 mg/kg/d) and captopril(30 mg/kg/d),and model group and normal group were given normal saline solution(2 mL/kg/d) all for 8 weeks.The pathological changes and fibrosis of myocardium were observed by applying HE and VG staining.The expressions of myocardial transforming growth factor β1(TGFβ1) and connective TGF(CTGF) were detected by using ELISA.Results The systolic pressure increased significantly in SHR(14 weeks old) compared with Wistar rats(14 weeks old,P0.01).After 8 weeks,the systolic pressure was significantly higher in the model group(22 weeks old) than that in the same group(14 weeks old,P0.05).The systolic pressure decreased significantly in the Baijili group than that in the model group(P0.01).The outcomes of VG staining showed that the model group(22 weeks old) had myocardial fibrosis and relieved in all medicinal groups similarly.The expressions of TGFβ1 and CTGF increased significantly in the model group compared with the normal group(P0.05).Compared with the model group,the expression of TGFβ1 was down-regulated in the captopril group(P0.05),and the levels of TGFβ1 and CTGF were significantly down-regulated in the Baijili group(P0.05).The levels of TGFβ1(P0
