摘  要:目的研究吴茱萸碱(Evo)对人胃腺癌(SGC-7901)细胞的作用,并初步探讨其作用机制。方法 MTT法测定Evo的抗肿瘤活性;电镜观察Evo对细胞形态学的影响;琼脂糖凝胶电泳检测Evo对细胞DNA电泳行为的影响;流式细胞技术检测Evo对SGC-7901细胞的抑制作用和细胞周期的影响。结果一定浓度范围内Evo对SGC-7901细胞的增殖有剂量、时间依赖性抑制作用;透射电镜下可见细胞核染色质边集、核碎裂及凋亡小体;一定浓度的Evo可使细胞基因组DNA电泳出现阶梯状条带;流式细胞仪检测Evo作用后,G0/G1期和S期细胞减少,大量细胞停滞在G2/M期,凋亡细胞增多。结论 Evo对SGC-7901细胞的抑制作用与诱导其凋亡使细胞停滞于G2/M期密切相关。Objective To study the effect of evodiamine(Evo) on human gastric adenocarcinoma cell line(SGC-7901) and discuss the mechanism.Methods The anti-tumor activity of Evo was determined by using MTT assay,and the influence of Evo on morphological changes was observed with the transmission electron microscope(TEM).The influence of Evo on the electrophoretic behavior of DNA was detected by using agarose gel electrophoresis,and the inhibitory effect and influence of Evo on SGC-7901 and cell cycle were detected by using flow cytometry.Results Evo,in a certain concentration,had a dose-time-dependent inhibitory effect on the proliferation of SGC-7901.The results of TEM showed nucleus chromatin margination,nuclear fragmentations and apoptotic bodies.Evo,in a certain concentration,brought cell genome DNA out ladder bands.The outcomes of flow cytometry showed that the cells of G0/G1 phase and S phase decreased,a lot of cells arrested in G2/M phase,and apoptotic cells increased.Conclusion The inhibitory and apoptosis-induced effects of Evo on SGC-7901 are closely related to that it can induce the apoptosis and bring the cells arrest in G2/M phase.
