摘 要:目的:探讨运动性低血色素形成过程中机体铁代谢的调节机制。方法:36只雄性Wistar大鼠随机分成安静对照组(control group,CG)和运动性低血色素组(exercise-induced hypochrosis group,EG),分别于运动后3周末、4周末和5周末取材,每次各取6只。RealTime-PCR检测大鼠肝HJV mRNA的表达;酶联免疫吸附法(ELISA)测血清Hepcidin。结果:1)与CG相比,大鼠Hb、RBC及Hct在运动后逐渐降低;2)与CG相比,大鼠的白细胞含量在运动4周后升高(P<0.05),5周后显著升高(P<0.01);3)运动3周后大鼠的肝HJVmRNA的表达和血清Hepcidin含量较对照组明显降低(P<0.05);运动4周后无明显变化(P>0.05);而运动5周后显著升高(P<0.01);结论:长时间大强度运动可引起机体的炎症反应和肝脏HJV的表达增强,进而提高Hepcidin的水平,这可能是运动性低血色素形成的重要原因之一。Objective:To discuss the mechanism of iron metabolism during the formation of exercise-induced hypochrosis.Method:36 male Wistar rats were randomly divided into two groups:control group(CG) and exercise-induced hypochrosis group(EG),and were sampled after 3,4 and 5-week running.The expression of hepatic hepcidin mRNA was examined by Real-Time PCR.The status of serum hepcidin was examined using Enzyme linked immunosor-bent assay(ELISA).Results:1) The level of hemoglobin(Hb),red blood cells(RBC),hematocrit(Hct) in EG was significantly decreased compared with CG.2) The content of WBC was significantly increased in 4-week EG and 5-week EG compared with 4-week CG and 5-week CG(P0.05 and P0.01).3) The HJV mRNA in liver and serum hepcidin of 3-week EG were significantly lower than that of the 3-week CG(P0.05).There was no significant changes of HJV mRNA and serum hepcidin between 4-week EG's and CG(P0.05).The HJV mRNA and serum hepcidin in 5-week EG were significantly higher than those in 5-week CG(P0.01).Conclusion:The log-term intensity exercise could increase inflammation and expression of hepatic HJV and in turn the expression of hepatic hepcidin mRNA was increased.It may be one of the most important reasons for the formation of the exercise-induced hypochrosis.
