摘  要:从赤子爱胜蚓(Eisenia foetida)匀浆液中提取出粗酶液,利用DNS法分析粗酶液中内切型纤维素酶(也称Cx酶)的活力.用刚果红染色法确定了赤子爱胜蚓含有3种内切型纤维素酶,将其分别称作Cx酶Ⅰ、Cx酶Ⅱ和Cx酶Ⅲ.经DEAE-Cellulose-52离子交换层析分离内切型纤维素酶,得到0.15~0.20mol/L NaCl洗脱峰含有活性组分.以制备电泳得到了Cx酶Ⅰ和Cx酶Ⅱ,并用SDS-PAGE测定它们的分子质量分别为33 400u和28 800u.用DNS法测得在粗酶液中内切型纤维素酶的最适反应温度为40℃,最适反应pH值为7.0.The crude enzyme solution from Eisenia foetida homogenate liquid was extracted,and the activity of endocellulases(also called Cx enzymes) was analyzed by DNS method.Three Cx enzymes were identified by Congo red staining,named as Cx enzyme Ⅰ,Cx enzyme Ⅱ and Cx enzyme Ⅲ respectively.DEAE-Cellulose-52 ion exchange chromatography were used to separate endocellulases,it was found that the effluent of 0.15~0.20 mol/L NaCl contained the active component.Cx enzyme Ⅰ and Cx enzyme Ⅱ were obtained by preparative electrophoresis,their molecular weights were 33 400 u and 28 800 u respectively,which determined by SDS-PAGE.The optimal temperature of endocellulases in the crude enzyme solution measured by DNS method was 40 ℃,and the optimal pH was pH 7.0.
