摘　　要：目的:改进SU11274的制备工艺。方法:以乙酰乙酸叔丁酯为起始原料通过8步反应合成了c-Met酪氨酸蛋白激酶抑制剂SU11274([(3Z)-N-(3-氯苯基)-3-({3,5-二甲基-4-[(4-甲基哌嗪-1-基)羰基]-1H-吡咯-2-基}亚甲基)-N-甲基-2-吲哚酮-5-磺酰胺])。结果和结论:目标产物结构经1H-NMR和ESI-MS确证,产率为56.1%。改进后的制备工艺反应条件温和,适合于实验室研究的小规模制备。Objective：To synthesize SU11274,an important small molecular tyrosine kinase inhibitor of c-Met,and optimize the preparing process.Methods： Using tert-butyl 3-oxobutanoate as starting material,（3Z）-N-（3-Chlorophenyl）-3-（{3,5-dimethyl-4-[（4-methyl-piperazin-1-yl） carbonyl]-1H-pyrrol-2-yl} methylene）-N-methyl-2-oxo-2,3-dihydro-1H-indole-5-sulfonamide,SU11274,was synthesized through 8 steps.Results and Conclusion： The structure of SU11274 was verified by 1H-NMR and ESI-MS.The improved process needs mild reaction condition,and is very suitable for lab-scale production.
