摘 要:目的:观察附子理中丸对脾阳虚证大鼠骨骼肌肌球蛋白ATP酶(M-ATP酶)活性的影响。方法:18窝新生大鼠,同窝9只匹配、体质量均衡随机分入9组;前5组不切除甲状腺为获得性脾阳虚;后4组切除甲状腺为发育性脾阳虚。第1d切除部分甲状腺,第42d切除肩胛骨间棕色脂肪,第56d开始高脂饲养,第70d开始用药防治,第98d取左侧腓肠肌,冰冻切片,M-ATP酶染色,形态计量单位体积相对总活性,t检验比较组间差别。结果:获得性脾阳虚中,模型组M-ATP酶活性比对照组升高26.10%(P<0.01),高脂组比模型组降低10.40%(P<0.01),凉治组比高脂组降低21.39%(P<0.01),温治组比高脂组升高34.89%(P<0.01)。发育性脾阳虚中,模型组M-ATP酶活性比对照组升高101.56%(P<0.01),高脂组比模型组降低19.98%(P<0.01),凉治组比高脂组升高8.01%(P<0.01),温治组比高脂组降低18.65%(P<0.01)。与获得性脾阳虚比较,性脾阳虚模型组、高脂组、凉治组和温治组分别升高59.85%(P<0.01)、42.76%(P<0.01)、96.14%(P<0.01)和18.29%(P<0.01)。结论:附子理中丸回调脾阳虚大鼠骨骼肌M-ATP酶活性,对发育性脾阳虚大鼠的作用效能大于获得性脾阳虚的作用效能。Objective: To observe the effect of Fuzi Lizhong Pills on the myosin ATPase(M-ATPase) activity of the skeletal muscle in rats with spleen yang deficient syndrome.Methods: Eighteen litters of rats were used,and nine littermates in each litter were bred sequentially and randomized into 9 groups.Five groups were not carried out thyroidectomy,the other four groups did.Thyroidectomy in hypothyroidism littermates was carried out with 131iodine to destroy partly thyroidal tissue at d1 after birth,the interscapular brown adipose tissue was removed at d42,littermates were fed with more cholesterol ester and triglyceride from d56,then administered intragastrically a decoction of medical herbs to promote or inhibit chronic thermogenesis from d70.At d98,gastrocnemius was taken from left leg of rats,then,the enzyme histochemistry for M-ATPase was carried out on the frozening sections pre-incubation at pH 10.4.The potency data were derives from the primary data.A t-test was used to evaluate the change between groups(P0.05) as the statistic significant.Results: In the euthyroid rats,compared with the control,the activity of M-ATPase in skeletal muscle increased by 26.10%(P0.01) in model rats.The activity in fat-diet rats decreased 10.40%(P0.01) than that in the model rats.The rats treated with Huanglian-Jiedu Decoction decreased the activity by 21.39%(P0.01) and the rats treated with Aconitum Lizhong Pill increased by 34.89%(P0.01) than that in the fat-diet rats.In the hypothyroid rats,compared with the control,the activity of M-ATPase in skeletal muscle increased by 101.56%(P0.01) in model rats;the activity in fat-diet rats decreased 19.98%(P0.01) than that in the model rats;the rats treated with Huanglian-Jiedu Decoction increased the activity by 8.01%(P0.01) and the rats treated with Fuzi Lizhong Pills decreased by 18.65%(P0.01) than that in the fat-diet rats.The activity potency of energy charge in the groups with hypothyroid was higher than that in the design-mated groups with
