摘 要:为弄清朱红橘红色突变体牛肉红金橘遗传的真实性,利用AFLP分子标记技术对朱红橘(Citrus reticulata Blanco)及其实生变异体牛肉红金橘进行了遗传鉴定。结果表明:从120对EcoRI/MseI酶切引物中筛选出具有多态性的引物7对(MCO5E04,MCO4E01,MC04E02,MC04E03,MC04E04,MC04E06,MCO5E02);对朱红橘及其突变体牛肉红金橘进行AFLP扩增,共扩增出648条带,其中,72条具有多态性,多态性比例占11.11%;应用NTSYSpc 2.1软件统计分析,品种(系)间的遗传距离为0.0215853。牛肉红金橘与朱红橘在DNA水平上确实存在差异,牛肉红金橘为遗传上稳定的变异体,具有成为新的红肉品种的遗传基础,可作为新型材料加以保护和利用。AFLP technique was used to determine DNA polymorphism of Zhuhongju and its variant line Niurou Hongjinju(Citrus reticulata Blanco) to study the genetic diversity of Zhuhongju(Citrus reticulata Blanco) and its red variant line.The results showed that seven primer pairs(MCO5-E04,MCO4-E01,MC04-E02,MC04-E03,MC04-E04,MC04-E06,MCO5-E02) were selected out of 120 pairs of primer combinations and 648 polymorphism bands were amplified,72 of which were polymorphic.Ratio of polymorphic bands was 11.11 %.The genetic distance between Zhuhongju and its variant line was 0.0215853 by NTSYSpc 2.1 cluster analysis.Niurouhongjinju was different with Zhuhongju at DNA level and was a genetically stable variant which had got genetic basis to be seleted as a new cultivar and should be protected and utilized as a new material.
