生物抗菌性空气过滤材料的制备及其抗菌效果评价

摘  要:目的制备新型生物抗菌性空气过滤材料,并进行抗菌效果评价,为研制新型抗菌过滤器等提供新材料。方法该研究选取安全性高的生物抗菌剂(硫酸鱼精蛋白、ε-聚赖氨酸、溶菌酶、硫酸多粘菌素、乳酸链球菌素)和常用的过滤材料玻璃纤维为基材,采用化学法对其进行表面修饰,从而将生物抗菌剂以共价键结合的方式固定在玻璃纤维上。同时考察固定时抗菌剂的浓度对固定后获得的抗菌滤材抗菌性能的影响,以及2种抗菌剂组合固定的抗菌效果。以大肠杆菌(8099)、金黄色葡萄球菌(ATCC 6538)、枯草芽孢杆菌(ATCC 9372)和噬菌体f2为模式微生物,采用液滴气相试验法对抗菌玻璃纤维进行抗菌效果评价。按照消毒技术规范的稳定性测试方法检测抗菌过滤材料的长效性。结果抗菌实验结果表明,硫酸鱼精蛋白、ε-聚赖氨酸和硫酸多粘菌素分别以55、1、mg/ml的质量浓度进行固定时,获得的抗菌滤材抗菌效果较理想,但是其对病毒的杀灭率小于99%。抗菌剂组合固定获得的抗菌过滤材料抗菌性能明显提高,以硫酸鱼精蛋白和甘氨酸、硫酸鱼精蛋白和ε-聚赖氨酸组合效果最好,其对4种模式微生物的抗菌率均达到99%以上。长效性测试结果表明这2种抗菌滤材在常温下的使用期限至少为2年。结论本研究制备了2种生物抗菌性玻璃纤维过滤材料,其对4种模式微生物的抗菌率均达到99%以上,且稳定性能良好,可长时间保存或使用。Objective To prepare a novel bio-antibacterial air filter material and evaluate its antibacterial efficiency,so as to provide the basis for developing new antibacterial filter.Methods Surface modifications of the glass fiber filter were performed via chemically modified procedure and the bio-antibacterial substances(protamine sulfate,ε-polylysin lysozyme,polymyxin sulfate,and nisin) were immobilized on it.Then the effect of different concentration of antibacterial agent and coupling of two kinds of antibacterial agent immobilized on the glass fiber was investigated.Escherichia coli(8099),Staphylococcus aureus(ATCC 6538),Bacillus subtilis(ATCC 9372) and E.coli phage f2 were selected as pattern microbe and liquid droplet method was used to test the antibacterial efficiency of antibacterial filters,then the longevity of antibacterial filter was tested according to the method of stability test in Technical Standard For Disinfection(2002 edition).Results When protamine sulfate,ε-polylysine and polymyxin sulfate of the concentration of 5,5,and 1 mg/ml respectively were immobilized,the antibacterial filters obtained had good antibacterial activity,but the anti-virus efficiency was less than 99%.The antibacterial filters immobilized with two kinds of antibacterial agents possessed better antibacterial activity.The combination of protamine sulfate and glycine,protamine sulfate and ε-polylysine got the best antibacterial efficiency(more than 99%).The filter material could be preserved or used at room temperature for two years.Conclusion Two kinds of antibacterial glass fiber filters were successfully prepared,the antibacterial efficiency of them against four model microorganisms were more than 99%,and the stability of them were also good.
