摘 要:目的调查广东人群CYP2C9基因第6外显子895A>G位点多态性情况。方法 PCR扩增跨越该位点的DNA片断,用VpaK11BI酶切PCR产物,通过PAGE-银染判断基因型。结果在受检的252例广东人中,共检出3例A895/G895杂合子,余者皆为野生型纯合子A895/A895。结论广东人群存在CYP2C9895G即CYP2C9*16等位基因,其频率为0.006。本研究为广东人群使用CYP2C9底物药提供了参考依据。Objective To investigate the polymorphism of A895G in exon 6 of CYP2C9 gene in Guangdong population. Methods The DNA fragments spanning the loci A895G in exon 6 of CYP2C9 gene were amplified by polymerase chain reaction,the PCR products were subjected to VpaK11BI digestion, then the genotypes were determined with polyaerylamide gel electrophoresis (PAGE) and sliver-staining. Results Three individuals were heterozygotes (A895/G895) in 252 individuals studied, others were wild-type homozygotes. Conclusion There was CYP2C9 895G namely CYP2C9 * 16 allele in Guangdong population, whose frequency was 0. 006. This study has provided some reference for the appropriate usage of drugs metabolized by CYP2C9.
