摘 要:针对红豆杉细胞生长相对缓慢和培养褐化问题,本文通过正交实验和均匀实验方法并采用人工神经网络技术对南方红豆杉细胞悬浮培养条件进行优化,使细胞的生长速率和比生长速率有较大提高,并考察了细胞活性随时间的动态变化.For solving the problem of slow growth rate of Taxus chinesis var. mairei cell suspension culture, the effects of different combination of sucrose, casamino acids,NAA,6-BA on plant cell suspension culture were optimized by experimental design of multi-factors orthogonal design. The growth rate and specific growth rate of Taxus chinesis var. mairei cells has been increased by the above method and the growth rate, cell density and specific growth rate reached 1. 05 g dry weight/L. d, 27. 6 g dry weight/L and 0. 154 I/L. d respectively, which have practical importance in large scale plant cell fermentation process. For preventing phenomenon of turned brown in Taxus chinesis var. mairei cell suspension culture,the effects of Ve,Vc,PVP on plant cell suspension culture were optimized by uniform design method combined with artificial neural network. It can be found that Ve and PVP had positive effect on antioxygenation and the optimal addition volume of Ve was 7 ml/L. Dynamic change of living-cell activity was examined by TTC method, it was shown that the change of living-cell activity did't keep pace with the change of cell weight in cell suspension culture and subculture should be in exponential phase for shortening lag hpase.
