摘 要:目的探讨糖尿病肾病(DN)早期大鼠肾组织中硫氧还蛋白相互作用蛋白(Txnip)表达的变化。方法 Wistar大鼠随机分成正常对照组(NC组)、糖尿病模型组(DM组)。腹腔注射链脲佐菌素诱导糖尿病模型,4周时取大鼠肾脏,HE染色观察大鼠肾组织的病理变化,免疫组化检测大鼠肾组织中Txnip的表达分布,Western blot和RT-PCR进一步检测Txnip蛋白和基因的表达。结果在DN早期,与NC组肾组织相比,DM组的肾小管和肾小球中均发现Txnip蛋白和基因的表达增加。结论 Txmp表达的变化在大鼠DN早期发挥重要作用。Objective To investigate expression of thioredoxin interacting protein (Txnip) in rat kidneys in the early stage of diabetic nephropathy. Methods Diabetic model rats were induced by intraperitoneal injection of STZ. Rats were divided into normal group and DM model group. In the fourth week after the STZ treatment, histopathological changes of rat kidneys were observed by HE staining, and Txnip was detected by immunohistochemical technology. Furthermore protein and mRNA expression of Txnip was analyzed by Western blot and RT-PCR separately. Results Immunohistochemical stain showed that Txnip protein located in the renal tubule and glomeruli of diabetic rats. The levels of Txnip protein and mRNA were significantly increased in DM group as compared with normal group. Conclusions Up-regulation of Txnip expression is associated with the early stage of diabetic nephropathy in rats.
