摘  要:目的通过研究吸烟对致敏大鼠气道组蛋白去乙酰化酶2(histone deacetylasese 2,HDAC2)及血清中白介素8(interleukin-8,IL-8)表达的影响,探讨吸烟支气管哮喘(简称哮喘)气道炎症的特点。方法雄性Wistar大鼠30只,随机分为对照组、致敏组和吸烟致敏组,每组10只。后两组应用卵白蛋白(OVA)致敏并长期(10周)吸入激发,制备哮喘模型。在激发第3周开始,将吸烟致敏组大鼠置于自制熏箱内进行被动吸烟。采用免疫组织化学法及双抗体夹心酶联免疫吸附试验法(ELISA)检测各组大鼠气道HDAC2蛋白及血清IL-8的表达。结果①吸烟致敏组气道HDAC2蛋白表达(0.339±0.050)明显低于致敏组(0.417±0.044)和对照组(0.472±0.047),差异有统计学意义(P值均〈0.05)。致敏组低于对照组,差异有统计学意义(P〈0.05)。②吸炯致敏组血清IL-8表达(63.04±7.06)ng/L明显高于致敏组(55.97±7.09)ng/L和对照组(47.25±4.94)ng/L,差异有统计学意义(P值均〈0.05)。致敏组高于对照组,差异有统计学意义(P〈0.05)。③HDAC2蛋白表达与IL-8表达呈负相关(r=-0.59,P〈0.05)。结论吸烟可使致敏大鼠气道HDAC2蛋白表达水平降低,血清IL-8表达水平增高,且二者呈负相关,提示吸炯可以进一步加重哮喘的气道炎症。Objective To study the effect of smoking on airway inflammation of bronchial asthma (asthma) and put forward experimental basis for the treatment of asthma smoking through researching the effect of smoking on the expressional levels of bistone deacetylase2 (HDAC2) protein and interleukin 8 (IL-8) in sensitized rats airway and serum. Methods 30 male Wistar rats were divided randomly into control group, sensitized group and smoking sensitized group. The rats of latter two groups were sensitized and challenged by ovalbumin to establish the asthmatic model. From the challenged third weeks, the smoking sensitized group rats were put in the self-made fumigating box to smoke passively. The expressions of HDAC2 protein and IL-8 in airway and serum were defected using immunohistochemistry and EI.ISA in all rats of three groups. Results (1) The expression of HDAC2 protein of smoking sensitized group (0. 339±0. 050) were significantly lower than those of the sensitized group (0. 417±0. 044) and the control group (0. 472±0. 047) (all P 〈0.05), so did the sensitized group and the control group,(P〈0.05). (2)TheexpressionofIL 8 of smoking sensitized group (63.04±7.06) ng/L were significantly higher than those of the sensitized group (55.97 ± 7.09) ng/L and the control group (47.25±4.94) ng/L. (all P 〈0.05), so did the sensitized group and the control group ( P 〈 0.05). (3) The expressions of HDAC2 protein were negatively correlated with the expression of IL-8 ( r = 0.59, P 〈 0.05). Conclusions Smoking can decrease the expression level of HDAC2 protein in sensitized rats airway and increase the expression level of IL- 8 in sensitized rat serum, there existe negative correlation between the expression of histone HDAC2 protein and IL -8, so smoking could aggravateasthma airway inflammation.
