摘 要:目的探讨RT-PCR扩增Sj26、Sj32和Sj14-3-3抗原编码基因用于慢性日本血吸虫病诊断的价值。方法从慢性日本血吸虫病患者血清中提取总RNA,用RT-PCR扩增Sj26、Sj32和Sj14-3-3抗原编码基因,以1.2%琼脂糖凝胶电泳进行鉴定,同时以华支睾吸虫病、卫氏并殖吸虫病、泡型包虫病、囊型包虫病、乙型肝炎、肺结核患者血清和健康人血清作为对照。结果 RT-PCR扩增出约400 bp的Sj14-3-3抗原编码基因片段,但未扩增出676 bp的Sj26和1 270bp的Sj32抗原编码基因片段。对照组呈阴性反应。结论 RT-PCR扩增Sj14-3-3抗原编码基因可用于慢性日本血吸虫病的基因诊断。Objective To investigate the diagnostic value of antigen genes Sj26,Sj32,and Sj14-3-3 amplified by RT-PCR for diagnosis of chronic schistosomiasis japonica.Methods Total RNA was extracted from sera of patients with chronic schistosomiasis japonica.Antigen genes Sj26,Sj32,and Sj14-3-3 were amplified by RT-PCR and identified by 1.2% agarose gel electrophoresis.The sera of patients with clonorchiasis sinensis,paragonimiasis westermani,alveolar echinococcosis,cystic echinococcosis,hepatitis B,or pulmonary tuberculosis and sera of healthy people served as a control.Results Sj14-3-3 antigen gene fragments of about 400 bp were obtained from total RNA by RT-PCR,but Sj26 and Sj32 antigen gene fragments of 676 bp and 1270 bp,respectively,were not obtained.Normal control groups were negative.Conclusion RT-PCR-amplified Sj14-3-3 antigen genes can be used in the genetic diagnosis of chronic schistosomiasis japonicum.
