摘  要:目的观察蒿甲醚体外抗多房棘球绦虫原头蚴作用,探讨治疗泡球蚴病新药物。方法将多房棘球绦虫原头蚴随机分为4组:蒿甲醚低剂量组(50μg/ml),蒿甲醚高剂量组(100μg/ml),阿苯达唑组(50μg/ml),空白对照组。每组设2个平行组,每组约含3 000个原头节,分别置含10 ml DMEM培养液的培养瓶中,每天观察1次,取样滴于玻片,用0.03%美蓝染色,显微镜下计数原头蚴约200个,计算死亡率。直至该组原头蚴全部死亡时培养结束,并对原头蚴进行超微结构观察。结果蒿甲醚50μg/ml和100μg/ml组不同作用时间原头蚴死亡率与空白对照组相比差异均有统计学意义(P<0.05);高、低剂量组原头蚴死亡率差异无统计学意义(P>0.05)。培养至第9 d时,蒿甲醚高剂量组原头蚴死亡率达100%,蒿甲醚低剂量组原头蚴死亡率93.28%,阿苯达唑组原头蚴死亡率99.03%,空白对照组原头蚴死亡率78.83%。结论蒿甲醚具有体外抗多房棘球绦虫原头蚴作用,且呈剂量、时间依赖。Objective To investigate the anti-protoscolex efficacy of artemether onEchinococcus multilocularis in vitro.Methods E.multilocularisprotoscoleces(EmPSC) were divided into four groups: a group subjected to a low dose of artemether(50 μg/ml),a group subjected to a high dose of artemether(100 μg/ml),a group subjected to albendazole(50 μg/ml),and a control group.Each group had 3,000 EmPSC that were cultured in DMEM culture media.EmPSC were dyed with 0.03% methylene blue and observed under a microscope.The mortality rate of EmPSC in each group was counted daily;in some groups,culturing was halted when EmPSC mortality reached 100%.Results There were significant differences(P0.05) in EmPSC mortality among the artemether 50 μg/ml group,artemether 100 μg/ml group,and the control group.There were no statistical differences(P0.05)between the artemether high-dose group and low-dose group.On day 9 of culturing,the artemether high-dose group had 100% mortality of EmPSC while the artemether low-dose group had 93.28% mortality,the albendazole group had 99.03% mortality,and the control group had 78.83% mortality.Conclusion Artemether was found to counter EmPSC in vitro and do so in a dose-and time-dependent manner.
