摘 要:目的探讨褪黑素(melatonin,MT)对HeLa细胞生长的影响及其对HeLa细胞中NF-κB蛋白表达的影响。方法 MTT法观察药物对HeLa细胞生长的影响,吖啶橙(acri-dine orange,AO)和溴化乙锭(ethidium bromide,EB)双染荧光显微镜观察MT对HeLa细胞形态学的影响,Western blot法检测HeLa细胞中的NF-κB蛋白水平。结果 MT作用72h,抑制HeLa的IC50为61.71 mg.L-1;MT 1 g.L-1作用1、2、3 d,细胞坏死随作用时间延长而增多;MT 0.2 g.L-1作用1,2、3 d以及MT 0.008、0.04和0.2 g.L-1作用3 d可下调HeLa细胞NF-κB蛋白水平,存在剂量依赖性与时间依赖性。结论 MT抑制HeLa细胞增殖可能与其下调NF-κB蛋白水平有关。Aim To investigate the anti-proliferative activities of melatonin(MT) and its influence on NF-κB level in HeLa cells. Methods The anti-prolifera-tive activities of MT were evaluated by MTT assay and AO/EB staining,and NF-κB level was observed by Western blot analysis. Results The IC50 on HeLa cells by MT for 72 h was 61. 71 mg . L -1. HeLa cells stained with AO/EB were damaged in a time-depend-ent manner by MT 1 g.L -1 for 1 d,2 d and 3 d. Asthe time goes on,more necrocytosis were seen and less apoptosis were observed. It was shown that MT could down-regulate NF-κB level in a time-dependent and a dose-dependent manner after treated with MT 0. 2 g.L -1 for 1 d,2 d and 3 d and with MT 0. 008,0. 04 and 0. 2 g.L -1 for 3 days. Conclusion MT could inhibit the proliferation of HeLa cells with a concomitant down-regulation of NF-κB level,which may be related to its antiproliferative activity.
