摘 要:【目的】对新疆近期发生的向日葵黑茎病进行病原菌分离鉴定,并对该菌进行RFLP分析。【方法】用常规分离培养法从田间采集的向日葵黑茎病病株获得致病菌,观察该菌的形态学特征;用PCR法对该菌ITS区进行扩增和测序,并与向日葵茎点霉黑茎病菌进行分子比对,明确二者的同源性;选取10种限制性内切酶对该菌的ITS区PCR产物进行RFLP分析。【结果】该菌的形态学特征为茎点霉属真菌,与向日葵茎点霉黑茎病菌的同源性达到99%以上,说明该致病菌为向日葵茎点霉黑茎病菌Phoma helianthi Taberosi。通过RFLP分析确定了该病菌的酶切位点。【结论】该致病菌的形态学和分子生物学鉴定结果表明,向日葵黑茎病由向日葵茎点霉黑茎病菌引起。[ Objective] The pathogen was isolated and identified which has caused sunflower black stem in Xinjiang recently. [ Method ] The pathogen was separated and cuhurated from the sunflower black stem' s plants which were collected in the fields by the traditional method. The fungus' morphology characteristics were observed, its district was analysed and amplified by PCR. The pathogen' s RFLP was analyzed by 10 kinds of different enzymes. [ Result] The morphological observation showed the pathogen belongs to Phoma. The homology of the pathogen and Phoma helianthi Taberosi reached 99%. The results identified that it belongs to Phoma helianthi Taberosi. [ Conclusion]The results indicated that the pathogen of sunflower black stem belongs to Phoma helianthi Taberosi.
