摘 要:【目的】探讨影响红腺忍冬外植体消毒灭菌和芽诱导效果的因子,为开发红腺忍冬组培快繁技术提供依据。【方法】以红腺忍冬带腋芽茎段为外植体,按不同灭菌时间(10、12、14、16min)、外植体取材季节(春、夏、秋、冬)、茎段直径大小(0.79、0.51、0.27cm)和处理部位(未木质化、半木质化、完全木质化)分别进行腋芽诱导。【结果】以0.1%升汞加吐温灭菌14min的外植体芽诱导率最高,为41.19%;未木质化茎段的芽诱导率最高,为41.36%;春季和秋季是比较理想的取材季节,外植体的芽诱导率分别为41.61%、39.42%;以平均直径为0.79cm的茎段进行初代培养,芽诱导效果最好,诱导率为41.81%,芽平均高度为2.35cm,平均叶片数为6.06张。【结论】选择未木质化、直径大小为0.79cm左右的茎段,在春季和秋季取外植体,以0.1%升汞加吐温灭菌14min,均可获得较理想的外植体芽诱导效果。[Objective]The effects of explant diameter, its sampling season and degree of lignification together with different time of sterilization have been studied on bud induction in Lonicera hypoglauca in order to provide a technique for its rapid propagation. [Method]The stems with different diameter (0.79, 0.51 and 0.27 cm), degree of lignification (nonlignified, semi-lignified and fully lignified) with axillary buds and sampled in different seasons (spring, summer, autumn and winter) were used as explants for bud induction using different sterilization times(10, 12, 14 and 16 min), respectively. [Result]The bud induction rate of explants sterilized with 0.1% HgCl2 and Twain for 14 min was found to be the highest (41.19%). Non-lignified stem, when used as explants, recorded highest bud induction rate (41.36%). The stems sampled in spring and autumn were found to be the better and recorded 41.61 and 39.62% induction rate, respectively. The explants of 0.79 cm average diameter recorded 41.81% induction rate which was better than the other ones. [Conclusion]The spring or autumn sampling for explants and selection of the non-lignified stems with approximately 0.79 cm diameter resulted in best bud induction rates when sterilized with 0.1% HgCl2 for 14 rain.
