摘 要:目的:研究白萝卜提取物(Raphanus sativus extract,Ecr)对在体小鼠胃排空及肠推进的影响及对家兔离体回肠平滑肌条收缩活动的影响。方法:通过水煮蒸发的方法,制备得到白萝卜提取物;将48只昆明小鼠随机分为白萝卜提取物高剂量(20 mL/kg)、中剂量(10 mL/kg)、低剂量(5 mL/kg)组及空白对照组(蒸馏水)4个组,每组12只,以胃肠内标记物墨汁在小鼠胃内色素相对残留率及肠推进比为指标,观察提取物对在体小鼠胃排空及肠推进的影响。进一步在离体条件下,以家兔回肠平滑肌肌条的张力和频率为指标,通过不同浓度的白萝卜提取物作用于家兔离体回肠平滑肌,观察提取物对离体回肠平滑肌收缩活动的影响。结果:白萝卜提取物在5 mL/kg、10 mL/kg、20 mL/kg剂量下都能够提高小鼠胃排空率和促进小肠的推进作用,与对照组相比,差异具有显著性;离体条件下,白萝卜提取物在0.1 g/L和0.2 g/L下可以提高家兔离体回肠平滑肌的张力和收缩频率,在0.05 g/L剂量下,家兔离体回肠平滑肌的张力、收缩频率无显著性变化。结论:白萝卜提取物对动物胃肠动力具促进作用,并存在一定的剂量-效应关系。Objective:To observe the effects of the Raphanus sativus extract(Ecr) on gastric emptying and small bowel peristalsis activity in mice in vivo and contraction activity of the smooth muscle of rabbit in vitro.Methods:The Raphanus sativus extract(Ecr) was obtained using rotary evaporation method.48 mice were randomly divided into four groups(12 in each group).The mice in experiment groups were administered with 5 ml/kg,10 ml/kg,20 ml/kg of Ecr with gavage respectively and the mice in the control group were given distillated water.By observing the residue rates of pigment in stomach and propulsive proportion of small intestine of ink in mice,the capacity of gastric emptying and small bowel peristalsis induced by Ecr were studied.Moreover,the samples of isolated ileum segment from rabbits were prepared,and the strain and frequency of the smooth muscle were measured before and after treatment of Ecr.Results: Compared with the control group,the Ecr could increase gastric emptying and small bowel peristalsis in mice in experiment groups with the dose of 5 mL/kg、10 mL/kg、20 mL/kg,and there were significant difference(P0.01).The Ecr could enhance the strain and frequency of the rabbit ileum smooth muscle in 0.1 mg/ml and 0.2 mg/ml group,(P0.05);while in 0.05 mg/ml group,the Ecr could not change the strain and frequency of the rabbit ileum smooth muscle in vitro,(P0.05).Conclusion: The Ecr can promote the gastrointestinal motility,and shows a dose-dependent relationship.
