柴芪益肝颗粒对小鼠肝移植瘤的作用

摘　　要：目的观察柴芪益肝颗粒(CQKL)对Hepa1-6肝癌细胞移植C57b l/6小鼠肝癌组织骨桥蛋白(OPN)及基质金属蛋白酶-2(MMP-2)表达的影响。方法构建C57b l/6小鼠肝癌模型,24只小鼠随机分为3组,观察组予柴芪益肝颗粒灌胃(273 mg/20 g),1次/d;空白对照组予生理盐水0.4 mL/20 g灌胃,1次/d;5-FU对照组采用腹腔注射5-FU[20 mg/(kg.d)];各组用药及处理均连续3周。观察各组抑瘤率、免疫组化法检测C57b l/6小鼠肝癌组织中OPN和MMP-2的表达。结果观察组和5-FU对照组的抑瘤率分别为38.52%和61.45%,与空白对照组比较,差异有统计学意义(P<0.01);观察组与5-FU对照组比较,差异有统计学意义(P<0.05),其作用弱于5-FU。柴芪益肝颗粒能抑制OPN和MMP-2的表达,与空白对照组比较差异有统计学意义(P<0.05)。结论柴芪益肝颗粒对肝移植瘤有一定的抑瘤及抗转移作用,其作用机制可能与肿瘤侵袭与转移密切相关的OPN和MMP-2表达有关,需要作进一步深入的研究。Objective To study the effects and mechanism of Chaiqiyigan Granula on growth and metastasis of transplanted human hepatocellular carcinoma in nude mice. Methods Twenty - four mice were selected to establish C57bl/6 animal model with the human hepatocellular carcinoma cell transplantation and were randomly divided into 3 groups： Chaiqiyigan Granula group [ 273 mg/（ 20 g ·d） ], 5 -FU control group[ 20 mg/（ kg ·d ） ] and NS control group. Chaiqiyigan Granula lavage, NS lavage and peritoneal injection of 5 - FU were performed in Chaiqiyigan Granula, NS control and 5 - FU conrol groups, respectively, for 3 weeks. Tumor inhibition rate, OPN and MMP - 2 expression were observed. Results The tumor inhibition rates in Chaiqiyigan granula and 5 - FU control groups were 38. 52% and 61.45%, respectively, with both significantly higher than that in NS control group （P 〈0. 01 ）. Significantly higher inhibition rate was observed in 5 - FU control group than that in Chaiqiyigan Granula group （ P 〈 0. 05 ）. Significant reduetion of OPN and MMP - 2 expression was also observed in Chaiqiyigan Granula group, comparing with that in NS control group （ P 〈 0. 05 ）. Conclusion Chaiqiyigan Granula can inhibit the growth and metastasis of hepatocellular carcinoma cells by regulating the expression of OPN and MMP - 2, which are associated with invasion and metastasis of hepatoeellular carcinoma cell.
