摘  要:分别采用平衡法和摇瓶法测定高、中、低纯度(71.0%、49.2%和25.4%)吴茱萸提取物中吴茱萸碱和吴茱萸次碱的溶解度及表观分配系数,采用HPLC法测定两者的浓度。高中低纯度吴茱提取物中吴茱萸碱和吴茱萸次碱均在PEG-400中溶解度最高,分别为8~14 mg/ml和13~19 mg/ml;异丙醇中的溶解度最低(均小于7 mg/ml)。吴茱萸碱和吴茱萸次碱在介质中的溶解度依次为PEG-400>Tween-80>RH-40>MCT>无水乙醇≈1,2-丙二醇≈1,3-丁二醇≈异丙醇。两者的表观分配系数的对数值(lgP)为0.6~1.2。吴茱萸提取物纯度越高,吴茱萸碱和吴茱萸次碱的溶解度越低;纯度与表观分配系数无相关性。pH对溶解度和表观分配系数均无明显影响。Solubility,apparent partition coefficient and concentration of evodiamine and rutaecarpine from Evodia rutaecarpa extracts with different purities(71.0%,49.2% and 25.4%)were determined by equilibrium,shake flask method and an HPLC method,respectively.Evodiamine and rutaecarpine with different purities were fairly dissolved in PEG-400 at 8-14 mg/ml and 13-19 mg/ml,but poorly in isopropanol at less than 7 mg/ml.Sequence of their solubility in solvents was PEG-400Tween-80RH-40MCT ethanol ≈ 1,2-propyleneglycol ≈ 1,3-butyleneglycol ≈ isopropanol.The range of logarithmes of apparent partition coefficient was 0.6-1.2.Solubility varied inversely with their purity while purity was irrelevant to their apparent partition coefficient.There was no effect of pH value on both solubility and apparent partition coefficient.
